Adiponectin gene ADIPOQ SNP associations with serum adiponectin in two female populations and effects of SNPs on promoter activity

Abstract: Adiponectin is an insulin sensitiser in muscle and liver, and low serum levels characterise obesity and insulin resistance. Eight tagging single nucleotide polymorphisms (tSNPs) in the ADIPOQ gene and promoter were selected, and association with serum adiponectin was tested, in two independent samples of Caucasian women: the Chingford Study (n = 808, mean age 62.8 ± 5.9 years) and Twins UK (n = 2,718, mean age 47.4 ± 12.6 years). In the Chingford cohort,

“…Two ADIPOQ promoter SNPs, rs17300539 (211391G.A) and rs266729 (211377C.G), have been investigated frequently in functional experiments (9,11,12,31) due to their strong reproducible correlations with circulating adiponectin and to the potential role of the promoter region in regulating gene transcriptional activity.…”

“…Among ADIPOQ variants identified by a candidate gene approach, the widely reported promoter SNP rs17300539 (211391 G.A) with potential biological function (16) was monomorphic in the CLHNS mothers. Four other candidate gene variants rs266729 (211377 C.G), rs2241766 (+45 T.G), rs1501299 (+276 G.T) and rs1063537 (11,13,16) were marginally associated with adiponectin (P ¼ 0.018, 0.0077, 0.059 and 0.017, respectively). The GWAS SNP rs3774261 (23) displayed significant association in the same direction in CLHNS (P ¼ 7.0 × 10 25 ).…”

“…AdipoQ rs1501259 T allele carriers were associated with high adiponectin levels in Italian population (14,30) and, just on the opposite, G allele carriers were associated with high adiponectin levels in Greek population (16) or significant associations were not found in a third group of studies (13). AdipoQ rs822391 T allele carriers were related to an increased risk for CVD in Korean volunteers (20). G=0.01 *1 subject failed to be genotyped for rs9939609 ** 1 subject failed to be genotyped for rs1501299 Table 4).…”

“…In addition, the area of the genome that harbors ADIPOQ has been identified by GWAS to be a susceptibility locus for risk of metabolic syndrome, T2DM and CVD (2). Studies have shown that ADIPOQ has some influence on adiponectin levels and plasma isoform levels, which are themselves related to modulation of the risk of developing T2DM and CVD (9).…”
