Background: Being obese is associated with both increased risk of developing multiple sclerosis (MS) and greater MS disease activity. Objectives: The objective of this study is to investigate levels and potential pathophysiologic contribution of serum adipose-hormones (adipokines) in pediatric-onset MS. Methods: Following a Luminex adipokine screen, adiponectin (APN) and its isoforms were quantified by enzyme-linked immunosorbent assay (ELISA) in 169 children with incident acquired demyelinating syndromes (ADS), prospectively ascertained as having either MS or other forms of inflammatory central nervous system (CNS) demyelination. The effect of recombinant APN and APN-containing sera was assessed on functional responses of normal human peripheral blood myeloid and T cells and on human CNS-derived microglia. Results: Compared to other cohorts, children with MS harbored higher serum APN levels, principally driven by higher levels of the low-molecular-weight isoform. Recombinant APN and pediatric MS serum-induced APN-dependent pro-inflammatory activation of CD14+ monocytes and of activated CD4+ and CD8+ T cells (both directly and indirectly through myeloid cells). APN induced human microglia activation while inhibiting their expression of molecules associated with quiescence. Conclusions: Elevated APN levels in children with MS may contribute to enhanced pro-inflammatory states of innate and adaptive peripheral immune responses and breach CNS-resident microglia quiescence, providing a plausible and potentially targetable mechanism by which APN contributes to MS disease activity.

Obesity causes low-grade chronic infl ammation. Adiponectin is an anti-infl ammatory cytokine. Increased waist circumference in obesity infl uences pro-infl ammatory and anti-infl ammatory cytokine levels which increase the risk of metabolic complications. This study was a cross-sectional study conducted at the Nutrition and Metabolic Diseases Clinic of Children's Hospital Dr. Soetomo, Surabaya. Subjects were chosen consecutively as many as 59 obese adolescents with inclusion criteria: obesity, aged 13-16 years, and having parents / guardians who were willing to follow the research and exclusion criteria: taking drugs, such as dyslipidemia, getting hormone therapy, suff ering from secondary obesity , suff ers from endocrine abnormalities, and has an infection or infl ammatory disease. Anthropometric examination was performed on subjects, including body weight, height, and waist circumference. Body Mass Index (BMI) is measured by weight calculation (kg) divided by height squared (meters) and plotted into the CDC 2000 curve. Obesity is established if a Body Mass Index/Age (BMI/A) >P95 is obtained according to age and sex on the CDC 2000 curve. Adiponectin levels are examined using venous serum with ELISA. The relationship between waist circumference and adiponectin levels was analyzed by Pearson correlation. 59 subjects were included in this study. The mean of waist circumference and adiponectin were 100.18 ± 10.63 cm and 7,84 ± 3,81 μg / ml. Statistic test showed that there is no correlation between waist circumference and adiponectin levels in obese adolescents (p>0,05). There is no correlation between waist circumference and adiponectin levels in obese adolescents. Factors that aff ect the correlation between waist circumference and adiponectin include race, diet and physical activity

Coronavirus disease-2019 (COVID-19) caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2; CoV2) is a deadly contagious infectious disease. For those who survived COVID-19, post-COVID cardiac damage poses a major threat for the progression of cardiomyopathy and heart failure. Currently, the number of COVID-related cases and deaths is increasing in Latin America, where a major COVID comorbidity is Chagas heart disease (caused by the parasite Trypanosoma cruzi). Here, we investigated the effect of T. cruzi infection on the pathogenesis and severity of CoV2 infection and, conversely, the effect of CoV2 infection on heart pathology during coinfection. We used transgenic human angiotensin-converting enzyme 2 (huACE2) mice infected with CoV2, T. cruzi, or coinfected with both in this study. Our study shows for the first time that white adipose tissue (WAT) serves as a reservoir for CoV2 and the persistence of CoV2 in WAT alters adipose tissue morphology and adipocyte physiology. Our data demonstrate a correlation between the loss of fat cells and the pulmonary adipogenic signaling and pathology in CoV2 infection. The viral load in the lungs is inversely proportional to the viral load in WAT, which differs between male and female mice. Our findings also suggest that adiponectin-PPAR signaling may differently regulate Chagas cardiomyopathy in coinfected males and females. We conclude that adipogenic signaling may play important roles in cardio-pulmonary pathogenesis during CoV2 infection and T. cruzi coinfection. The levels of adiponectin isomers differ between male and female mice during CoV2 infection and coinfection with T. cruzi, which may differently regulate inflammation, viral load, and pathology in the lungs of both the sexes. Our findings are in line with other clinical observations that reported that males are more susceptible to COVID-19 than females and suffer greater pulmonary damage.
