Adiponectin potentiates the acute effects of leptin in arcuate Pomc neurons

Abstract: ObjectiveAdiponectin receptors (AdipoRs) are located on neurons of the hypothalamus involved in metabolic regulation – including arcuate proopiomelanocortin (Pomc) and Neuropeptide Y/Agouti-related peptide (NPY/AgRP) neurons. AdipoRs play a critical role in regulating glucose and fatty acid metabolism by initiating several signaling cascades overlapping with Leptin receptors (LepRs). However, the mechanism by which adiponectin regulates cellular activity in the brain remains undefined.MethodsIn order to resolv… Show more

“…Similar to previous reports (Hill et al, 2008; Sohn et al, 2011), Pomc neurons that express LepRs were targeted from Pomc -hrGFP∷ LepR -cre-tdtomato (PLT) mice (Sohn et al, 2011; Sun et al, 2016; Williams et al, 2014) to test the acute cellular effects of leptin (Figures 3A to 3E). Leptin (100 nM) depolarized 70.6 % (12 out of 17) of wildtype Pomc neurons that express LepRs by 6.7 ± 0.5 mV (n=12, Figures 3F, 3J and supplemental table 1).…”

“…Slice preparation and whole-cell recordings. Brain slices were prepared from young adult male mice (7-9 weeks old) as previously described (Sohn and Williams, 2012;Williams et al, 2014;Sun et al, 2016;Gao et al, 2017). Briefly, male mice were deeply anaesthetized with 7% chloral hydrate (i.p.)…”

“…Leptin binds to the leptin receptor (Ob-Rb) in the ARC [9195] and stimulates the cellular activity of POMC neurons while inhibiting the cellular activity of NPY/AgRP neurons [33]. Leptin or leptin receptor -deficient rodents and humans are obese due to hyperphagia and reduced energy expenditure [91,96].…”
