Adjuvant effect of Bordetella pertussis vaccine to sheep erythrocytes: enhancement of antibody formation by using subcutaneous administration of adjuvant and antigen

Abstract: The subcutaneous route (s.c.) was used to study the adjuvant effect of Bordetella pertussis vaccine (PV) on the primary antibody response to sheep erythrocytes. The reasons for using the s.c. route are discussed. PV, besides enhancing the hemagglutinin response, also markedly increased the number of plaqueforming cells in the draining lymph nodes. A heated preparation of PV was tested and found to possess significant adjuvant activity. Interestingly, the enhancement occurred in the absence of marked enlargemen… Show more

“…The results in Table 3 are representative of three series of experiments and show that higher interferon activity was consistently observed in Lytl +depleted cultures, whereas Lyt2+ depletion resulted in lowered interferon activity. DISCUSSION B. pertussis cells have been reported to enhance antibody responsiveness to a variety of unrelated antigens (5,13,30). However, the best adjuvant effect is obtained when B. pertussis is injected simultaneously with antigen (5,30).…”

“…DISCUSSION B. pertussis cells have been reported to enhance antibody responsiveness to a variety of unrelated antigens (5,13,30). However, the best adjuvant effect is obtained when B. pertussis is injected simultaneously with antigen (5,30). If administered a few days before or after antigen, B. pertussis may actually suppress the response.…”

“…As with PV, the adjuvanticity of PS was completely eliminated by heat. DISCUSSION Whereas the enhancing effect of PV on the humoral antibody response to SRBC has been extensively studied (3,5,8,18,19), the ability of PV to enhance the CMI response to SRBC has received virtually no attention. In the present report, the adjuvant effect of PV on the CMI response to SRBC was studied.…”

“…However, the role of CMI in tumor rejection and inhibition of viral replication in these investigations was not substantiated. In a previous publication (19), we reported on an in vivo model of studying the adjuvant effect of PV on the antibody response to sheep erythrocytes (SRBC) in which we used exclusively the subcutaneous (s.c.) route of administration of adjuvant and antigen. Also, we have reported (18) on the adjuvant effect of PV on the in vitro antibody response to SRBC with normal spleen cells, studied through the Mishell-Dutton immunization technique.…”
