“…The results of the present study showed significantly reduced levels of IL-6, IL-8 and TNF-α in patients receiving vonoprazan fumarate tablets vs. those without, suggesting that vonoprazan fumarate tablets effectively alleviate clinical symptoms, promote esophageal mucosal healing and reduce inflammation in the gastric body and sinus of RRE patients. However, whether vonoprazan fumarate tablets have anti-inflammatory effects by non-acid-dependent mechanisms remains to be verified [22] .…”

“…Other studies have shown that exogenous hypergastrinemia stimulates GE, associated with increased motilin secretion in duodenal acidification [25]. However, a recent study has shown that a 2-week administration of vonoprazan, a newly released P-CAB in Japan, induced hypergastrinemia but did not demonstrate a significant correlation with GE [12]. While this study reported a decrease in liquid GE with vonoprazan, it is important to note that the measurement of GE was performed using liquid food instead of solid food, and the observation period was limited to post-prandial 120 min using breath tests, indicating potential limitations in its methodology.…”

“…This biological activity is demonstrated that the first day of 24-h intragastric pH monitoring after administration of tegoprazan 50mg is >4 for 76% of the time and this degree of acid suppression is virtually identical to that achieved (80%) with 14 days of administration [10,11]. In a recent study investigating vonoprazan's effect on GE, serum gastrin levels increased to over 500 pg/dL after 2 weeks of treatment, yet no association with gastric emptying was observed [12]. Furthermore, unlike vonoprazan, tegoprazan induces hypergastrinemia at levels similar to PPIs.…”

“…Low-grade inflammation as shown by eosinophil and mast cell infiltration of the duodenum is associated with functional dyspepsia [1,18] and acid suppression by VPZ may attenuate low-grade inflammation of the duodenum. A recent Japanese study revealed that VPZ delayed gastric emptying but increased ghrelin levels in healthy male subjects [22]. Vonoprazan surely improved dyspepsia symptoms as much as ACO did, and therefore delayed gastric emptying due to VPZ may be compensated by elevating ghrelin levels that can improve dyspepsia symptoms.…”
