Advanced Glycation End Product (AGE)-Receptor for AGE (RAGE) Signaling and Up-regulation of Egr-1 in Hypoxic Macrophages

Abstract: Receptor for advanced glycation end product (RAGE)-dependent signaling has been implicated in ischemia/reperfusion injury in the heart, lung, liver, and brain. Because macrophages contribute to vascular perturbation and tissue injury in hypoxic settings, we tested the hypothesis that RAGE regulates early growth response-1 (Egr-1) expression in hypoxia-exposed macrophages. Molecular analysis, including silencing of RAGE, or blockade of RAGE with sRAGE (the extracellular ligandbinding domain of RAGE), anti-RAGE … Show more

“…Although RAGE signaling via p38 MAPK and NF-B has recently been proposed in diverse tissue types under various conditions (19,40,54), such signaling paradigms have not been elucidated in AMs exposed to tobacco smoke. Importantly, the signaling kinetics presented in the current research are similar to those presented by Zhang et al (53) in that p38 MAPK activation peaked at 30 min and NF-B activation peaked at 4 h following exposure of RAW234.7 to AGEs.…”

“…However, RAGE-blocking antibody attenuated such effects, especially in HSCs treated with higher concentration. Soluble form of RAGE (sRAGE) binds RAGE ligands, thereby preventing downstream signaling and damage [28]. Next, sRAGE was used to block AGE-RAGE signaling.…”

“…The process of protein glycation by glucose. addition, RAGE induces the activation of signaling pathways such as nuclear factor of activated T-cells (NF-AT) [21], mitogen-activated protein kinase and extracellular signal-regulated kinase signaling [22], cAMP response element-binding factor [23], and c-Jun Nterminal kinase signaling [24]. The diversity of signaling cascades identified in RAGE-mediated cellular signaling suggests that there is no any single mode of RAGE activation by different RAGE ligands and calls for our further investigation.…”
