The ligule ultrastructure of the tidal swamp rice (Oryza sativa) landraces of South Kalimantan, Indonesia, and their genetic diversity and relationship. Biodiversitas 22: 5280-5285. In-depth characterization of rice landrace is needed in supporting the future conservation and crop breeding programs. Tidal swamp rice (Oryza sativa L.), belonging to this landrace, provided useful agronomic traits for both programs. In this study, the nine the tidal swamp rice ligule samples, mainly from South Kalimantan, Indonesia, were observed ultrastructurally using the SEM method. The diversity and relationships of this landrace based on the ligule ultrastructure were also determined using PCA and UPGMA methods. Generally the tidal swamp rice showed a unique ultrastructure of their ligule, composed of the large trichomes (prickle hair), small trichomes (micro hair), stomata-like structure, and silica cells, as well as the papillae that line along the veins. In this case, we found four patterns of the prickle hair on the ligule's abaxial surface, which has never been reported previously, namely scattered randomly, lined up on the vein, grid-step-like pattern, and unpatterned. The results of PCA and UPGMA showed that this landrace grouped into the same main three clusters, where Sardani (an outgroup) was included in the second cluster and joined with other cultivars from South Kalimantan. This information may be useful for the development of taxonomy and plant structures, and plant genetics and conservation.

Despite the almost universal acceptance of the phrase “you are what you eat,” investment in understanding diet-based nutrition to address human health has been dwarfed compared to that for medicine-based interventions. Moreover, traditional breeding has focused on yield to the detriment of nutritional quality, meaning that although caloric content has remained high, the incidence of nutritional deficiencies and accompanying diseases (so-called hidden hunger) has risen dramatically. We review how genome sequencing coupled with metabolomics can facilitate the screening of genebank collections in the search for superior alleles related to the nutritional quality of crops. We argue that the first examples are very promising, suggesting that this approach could benefit broader ranges of crops and compounds with known relevance for human health. We argue that this represents an approach complementary to metabolic engineering by transgenesis or gene editing that could be used to reverse some of the losses incurred through a recent focus on breeding for yield, although we caution that ensuring such approaches are not (re)introducing antinutrients is also necessary. Furthermore, deploying effective postharvest processing technologies to increase the density of nutrients in the food matrix and improve palatability remains an attractive alternative. Rice seeds at each stage of ripening are characterized by a certain state, structure and moisture. Such properties of rice as germination, nature, quality and duration of storage depend on moisture content. Biochemical processes in the grain do not end at the filling stage, but continue for some post-harvest period. To safely preserve the rice gene pool, it is necessary to clarify the effect of seed maturity and storage time on seed quality. The varietal characteristics of rice seeds were studied using 8 Kuban varieties harvested on days 35 and 45 after flowering. The structural-phase state of freshly harvested grain and after 5, 10 days of ripening in natural conditions at room temperatures, and then the dynamics of sowing qualities after 1, 6 and 10 months of storage are discussed. The following parameters were studied: moisture, germination, length of the seedling and seed root, mass of 1000 grains. Differences in the moisture yield of grain of different degrees of maturity, varieties with different types of grain are shown. The results of the evaluation of rice physico-chemical indicators at the end of post-harvest ripening are presented. Varieties with slow moisture yield during ripening were distinguished by increased grain size: Yakhont, Rubin, Thaibonne and Zlata. The germination of seeds increased as they ripened, compared with freshly harvested ones, after a month it increased by almost 50%, and reached its maximum value by 6 months of storage, after 10 months, on average, germination for the group of varieties has already decreased by 6%. The stage of seed harvesting significantly influenced the germination rates.
