Organ-on-a-chip devices are powerful modeling systems that allow researchers to recapitulate the in vivo structures of organs as well as the physiological conditions those tissues are subject to. These devices are useful tools in modeling not only the behavior of a healthy organ but also in modeling disease pathology or the effects of specific drugs. The incorporation of fluidic flow is of great significance in these devices due to the important roles of physiological fluid flows in vivo. Recent developments in the field have led to the production of vascularized organ-on-a-chip devices, which can more accurately reproduce the conditions observed in vivo by recapitulating the vasculature of the organ concerned. This review paper will provide a brief overview of the history of organ-on-a-chip devices, before discussing developments in the production of vascularized organs-on-chips, and the implications these developments hold for the future of the field.

The vasculature is an integral unit of the tissue microenvironment due to providing nutrients and oxygen to surrounding cells. Therefore, pro-angiogenic biomaterials have the potential to improve the success of a wide range of medical therapies, including tissue engineering, wound healing, and drug delivery. Herein, we decellularized bovine spinal cord meninges with Triton X-100 and digested them with pepsin to obtain a hydrogel (MeninGEL). The cryogel form of the MeninGEL was also prepared by lyophilization process (named as MeninRIX). DNA content analysis showed that the nuclear content was significantly reduced by 98.6% after decellularization process. Furthermore, the effect of decellularization on extracellular matrix components was investigated with glycosaminoglycan (GAG) and hydroxyproline (HYP) content analyses. Tensile, compression, and suture retention tests were performed to elucidate the mechanical properties. The physiological degradation behavior of the bioscaffolds was investigated by hydrolytically. Both MeninGEL and MeninRIX have good biocompatibility and pro-angiogenic properties, as proved by the Chick Chorioallantoic Membrane (CAM) assay. Moreover, SEM and histological analyses indicated cellular migration, attachment, and dynamism on the bioscaffolds’ surfaces. On the basis of these data, MeninGEL and MeninRIX are pro-angiogenic structures and have adequate mechanical properties, which makes them promising candidates for soft regenerative medicine applications.

Pro-angiogenic photo-crosslinked silk fibroin hydrogel: a potential candidate for repairing alveolar bone defects

Objective: This study aimed to develop a pro-angiogenic hydrogel with in situ gelation ability for alveolar bone defects repair. Methodology: Silk fibroin was chemically modified by Glycidyl Methacrylate (GMA), which was evaluated by proton nuclear magnetic resonance (1H-NMR). Then, the photo-crosslinking ability of the modified silk fibroin was assessed. Scratch and transwell-based migration assays were conducted to investigate the effect of the photo-crosslinked silk fibroin hydrogel on the migration of human umbilical vein endothelial cells (HUVECs). In vitro angiogenesis was conducted to examine whether the photo-crosslinked silk fibroin hydrogel would affect the tube formation ability of HUVECs. Finally, subcutaneous implantation experiments were conducted to further examine the pro-angiogenic ability of the photo-crosslinked silk fibroin hydrogel, in which the CD31 and α-smooth muscle actin (α-SMA) were stained to assess neovascularization. The tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were also stained to evaluate inflammatory responses after implantation. Results: GMA successfully modified the silk fibroin, which we verified by our 1H-NMR and in vitro photo-crosslinking experiment. Scratch and transwell-based migration assays proved that the photo-crosslinked silk fibroin hydrogel promoted HUVEC migration. The hydrogel also enhanced the tube formation of HUVECs in similar rates to Matrigel ® . After subcutaneous implantation in rats for one week, the hydrogel enhanced neovascularization without triggering inflammatory responses. Conclusion: This study found that photo-crosslinked silk fibroin hydrogel showed pro-angiogenic and inflammation inhibitory abilities. Its photo-crosslinking ability makes it suitable for matching irregular alveolar bone defects. Thus, the photo-crosslinkable silk fibroin-derived hydrogel is a potential candidate for constructing scaffolds for alveolar bone regeneration.
