Nanoparticles synthesis through biological mediated methods with a particular focus on the processes mediated by fungi and algae is discussed, which systematically reviews nanoparticle characterization, composition, synthesis methods, and, lastly but not least, the applications of NPs across five different categories to provide a reference for future research. Most traditional methods to generate nanoparticles have certain limitations, like the toxicity of precursor materials, the need for high-temperature management, and the high cost of synthesis, which ultimately hinders their utility in sectors. Greener synthesis through fungus and algae done through bioreduction by biomolecules or enzymes present in them is low-energy, low-cost, and needs a low-temperature environment, providing a unique technique for the manufacture of various metallic nanoparticles utilized in an array of industries and healthcare.

The rapid advancement of nanomedicine presents novel alternatives that have the potential to transform health care. Targeted drug delivery as well as the synthesis of nanocarriers is a growing discipline that has been intensively researched to reduce the complexity of present medicines in a variety of diseases and to develop new treatment and diagnostic techniques. There are several designed nanomaterials used as a delivery system such as liposomes, micelles, dendrimers, polymers, carbon-based materials, and many other substances, which deliver the drug moiety directly into its targeted body area reducing toxic effect of conventional drug delivery, thus reducing the amount of drug required for therapeutic efficacy and offering many more advantages. Currently, these are used in many applications, including cancer treatment, imaging contrast agents, and biomarker detection and so on. This review provides a comprehensive update in the field of targeted nano-based drug delivery systems, by conducting a thorough examination of the drug synthesis, types, targets, and application of nanomedicines in improving the therapeutic efficiency.

Hsp70‐Targeting and Size‐Tunable Nanoparticles Combine with PD‐1 Checkpoint Blockade to Treat Glioma

Invasive glioma usually disrupts the integrity of the blood‐brain barrier (BBB), making the delivery of nanodrugs across the BBB possible, but sufficient targeting ability is still avidly needed to improve drug accumulation in glioma. Membrane‐bound heat shock protein 70 (Hsp70) is expressed on the membrane of glioma cells rather than adjacent normal cells, therefore it can serve as a specific glioma target. Meanwhile, prolonging the retention in tumors is important for active‐targeting nanoparticles to overcome receptor‐binding barriers. Herein, the Hsp70‐targeting and acid‐triggered self‐assembled gold nanoparticles (D‐A‐DA/TPP) are proposed to realize selective delivery of doxorubicin (DOX) to glioma. In the weakly acidic glioma matrix, D‐A‐DA/TPP formed aggregates to prolong retention, improve receptor‐binding efficiency and facilitate acid‐responsive DOX release. DOX accumulation in glioma induced immunogenic cell death (ICD) to promote antigen presentation. Meanwhile, combination with the PD‐1 checkpoint blockade further activate T cells and provokes robust anti‐tumor immunity. The results showed that D‐A‐DA/TPP can induce more glioma apoptosis. Furthermore, in vivo studies indicated D‐A‐DA/TPP plus PD‐1 checkpoint blockade significantly improved median survival time. This study offeres a potential nanocarrier combining size‐tunable strategy with active targeting ability to increase drug enrichment in glioma and synergizes with PD‐1 checkpoint blockade to achieve chemo‐immunotherapy.
