Aeroallergen‐induced dyspnea in freely moving guinea pigs: quantitative measurement by bias flow ventilated whole body plethysmography

Abstract: Guinea pigs were sensitized and boostered with i.p. injections of ovalbumin (OA) 10 micrograms + Al(OH)3 100 mg. Thirteen days after the last injection animals (800-1100 g) were placed in bias flow ventilated whole body plethysmographs and allowed to stabilize for 2 h. Lung function was recorded for up to 2 h before and 5 h after aeroallergen challenge (OA 20 mg/ml, 60 s, 20 psi) by a noninvasive pulmonary analyzer for unrestrained rodents. Aeroallergen challenge produced immediate dyspnea and gasping (peaking… Show more

“…Therefore, in this study the Penh is considered as a total lung resistance index (iRL), as has been proposed by others and us (Allen et al 1995;Chand et al 1993;Hamelmann et al 1997;Sommer et al 1998).…”

“…The plethysmographic system has been previously described in detail (Chand et al 1993;Hamelmann et al 1997). Briefly, the pressure inside the chamber was measured by a differential pressure transducer (SCXL004DN; Sensym, Milpitas, Calif., USA) connected to a preamplifier, and continuously monitored through a software (Buxco Biosystem XA v1.1) in a microcomputer.…”

“…In 'f .,C, .ir a first one, 5-HT was administered i.v. to anesthetized mice (5,18), and in the second, methacholine was administered by aerosol to awake animals (19). This is important since bronchoconstrictor responses may differ according to the route of administration of the agonists, which determines the first line cell species that is encountered.…”

“…Indeed, IL-5 was present in the BALF and serum of the BP2 mice after antigen challenge and the anti-IL-5 antibody TRFK-5 suppressed altogether eosinophilia in airways and tissues and BHR. In allergen-challenged guinea pigs, anti-IL-5 antibody also suppressed airway eosinophilia (26) and BHR (27,28). CD4+ T lymphocytes that migrate into the lungs after antigen challenge (29) are responsible for the local production of IL-4 and IL-5, which are important in B-and T-lymphocyte development and function and in eosinophil activation and differentiation (30).…”

“…The potential for adverse effects on respiration or motor activity/sedation were assessed using whole-body plethysmography and rotarod test in rats. 119,120 Effects on respiration were quantified by the measurement of respiratory rate, tidal volume, peak inspiratory flow, peak expiratory flow, inspiration time and expiration time. At the maximum test dose tested of 16 lg/kg IV, which was 3Á2-fold the analgesic ED50 dose in the tail-flick test, cebranopadol did not produce any significant changes in the measured respiratory parameters or on In vitro human binding assay results motor coordination.…”
