Affimers and nanobodies as molecular probes and their applications in imaging

Abstract: Antibodies are the most widely used, traditional tool for labelling molecules in cells. In the past five to ten years, many new labelling tools have been developed with significant advantages over the traditional antibody. Here, we focus on nanobodies and the non-antibody binding scaffold proteins called Affimers. We explain how they are generated, selected and produced, and we describe how their small size, high binding affinity and specificity provides them with many advantages compared to antibodies. Of par… Show more

“…The structure in VNARs and the characteristics of the composition of the individual Loop loops allow them to recognize more hidden epitopes, and therefore VNARs may play an unexpected role in development studies of OGT and O-GlcNAcylation. Conventional monoclonal antibodies are limited in their in-depth application by their large size, complex structure and sensitivity to extreme environmental temperatures ( 42 ). Shark VNAR is better able to penetrate tissues and dense structures and penetrate deeper into the target protein.…”

“…Affimers could act as molecular probes to interrogate the interactions between receptor and ligand, or could themselves act as template biopharmaceuticals with beneficial effects on atherosclerotic plaque formation. To date, the majority of applications for Affimers have been as probes to support high-resolution imaging ( 51 , 52 ) or as antibody replacements in diagnostics and biosensor development ( 53 , 54 , 55 ). However, they have also proven to be powerful tools in the study of protein-protein interactions ( 56 , 57 ), with the capability to discriminate between very closely related proteins ( 58 ).…”

“…Affibodies can be Affimers. Affimers are scaffolding proteins that are not antibodies but are capable of binding to other molecules with high affinity and specificity [238]. Affimers are obtained using the phage display method, in which those that have the highest affinity for the analyte of interest are selected from a library of random protein sequences [239].…”
