Age‐dependency of glycosaminoglycan content in lumbar discs: A 3t gagcEST study

Abstract: We show a decreased gagCEST effect likely corresponding to decreasing glycosaminoglycans with aging. Hence, age-matched analysis of gagCEST imaging may be necessary in future studies.

“…The residual signal normalized to the reference image as a function of the offset frequencies (z-spectrum) can be utilized for the evaluation of GAG CEST effect 17 . GagCEST values were determined using the MTR asym value in the frequency range from 0.9 to 1.9 ppm, which comprises the chemical exchange resonances of GAG hydroxyl protons 14 .…”

“…Data analysis methods and calculations of gagCEST measurements were used according the work of Müller-Lutz et al 17 For reliability assessment, the analysis was repeated by the latter and by a second observer (AML) (medical physicist with four years of segmenting experience in musculoskeletal imaging).…”

“…179,181 GagCEST has also been applied to study nondegenerative lumbar intervertebral discs 182 and may differentiate between the nucleus pulyposus (NP) and annulus fibrosus (AF) of these discs. [183][184][185][186] GagCEST technology, similar to the APTw approach, is based on MTR asym analysis and optimization of the process, especially with respect to the correction of B 0 and B 1 inhomogeneities and removal of interference by NOEs, is still ongoing. Applications to larger study cohorts are needed to convert this into a clinically viable technique.…”
