Age-dependent stability of nucleoli and global DNA methylation level in spermatocytes of the domestic horse (Equus caballus)

Abstract: The aim of the study was to determine the number and shape of nucleoli during meiosis in cells of the domestic horse. In addition, the level of global DNA methylation was determined using a quantitative technique for measuring the relative level of DNA methylation, modelled on an immunoenzymatic assay. The research was carried out on stallions belonging to two age groups (2 and 7 yr). In the cells of the 2-yr-old animals, the nucleoli were mainly of a regular shape and no fragmented nucleoli were observed. The… Show more

“…In the present study, an experimental technique of staining spermatozoa with silver nitrate colloid solution was used. As silver nitrate is an alkaline dye, it is mainly used to identify acidic chromatin proteins and the chromatin of nucleolus organizer regions in mitotic chromosomes [ 57 60 ] and nucleoli during meiosis [ 61 , 62 ] The basic methodology has been modified in our Department (Department of Animal Genetics and Horse Breeding, University of Natural Sciences and Humanities in Siedlce) and successfully used to identify morphological details of the spermatozoa either from fresh, frozen or fixed semen sample of mammals [ 42 44 , 63 , 64 ], birds [ 47 ] and insects. [ 36 ]…”

“…According to published data, the presence of a tetraploid karyotype leads to the cessation of division, acceleration of cellular aging, and apoptosis (1). Changes in the amount of genetic material during cultivation can lead to abnormal functioning of the cell genome and negative effects in the cell, including oncogenic transformation (5).…”

“…In addition to the main livestock species discussed above, some epigenetics-related data have been reported in other livestock species. For instance, DNA methylation was predicted to be an age-dependent process in domestic horses ( Andraszek et al, 2016 ). Different from the generally methylated mammalian genomes, honeybees have unique methylation patterns that concentrate in gene bodies and are associated with gene expression ( Wedd and Maleszka, 2016 ; Harris et al, 2019 ).…”
