Accumulation of oxidative stress-induced damage in brain tissue plays an important role in the pathogenesis of normal aging and neurodegenerative diseases. Neuronal oxidative damage typically increases with age in humans, and also in the invertebrate and vertebrate model species most commonly used in aging research. By use of quantitative immunohistochemistry and Western blot, we show that this aspect of brain senescence is largely decoupled from chronological age in the honey bee (Apis mellifera). The bee is a eusocial insect characterized by the presence of a reproductive queen caste and a caste of functionally sterile female workers that performs various alloparental tasks such as nursing and foraging. We studied patterns of oxidative nitration and carbonylation damage in the brain of worker bees that performed nurse tasks as 8-and 200-day-olds and foraging tasks as 20-and 200-day-olds. In addition, we examined 180-day-old diutinus bees, a stress-resistant temporal worker form that survives unfavorable periods. Our results indicate that nitration damage occurs only at low levels in vivo, but that a 60-kDa protein from honey bee brain is selectively nitrated by peroxynitrite in vitro. Oxidative carbonylation is present at varying levels in the visual and chemosensory neuropiles of worker bees, and this inter-individual variation is better explained by social role than by chronological age.

-Oxidative stress is defined as a disturbance in the balance between the production of reactive oxygen species and antioxidant defences. We measured total antioxidant capacity (TAC) in honeybee haemolymph and seminal plasma and analysed TAC of haemolymph in relation to age and exposure to pesticide. TAC of haemolymph increased with age of bees (1.18 vs 1.97 mM of (±)-6-hydroxy-2,5,7,8-tetramethylchromane-2-carboxylic acid (Trolox) for 1-and 30-day-old bees, respectively, P ≤0.05). Exposure to imidacloprid (IMD) affected TAC of haemolymph of 1-day-old but not 30-day-old honeybees. TAC in haemolymph of 1-day-old bees was lower in treatments with the addition of 5 and 200 ppb IMD (1.57-1.46 mM of Trolox in treated bees compared with 2.37 mM of Trolox in controls; P ≤0.05). In conclusion, antioxidant protection of honeybees seems to be related to age and may be disturbed by exposure to IMD. Older bees with higher antioxidant protection seem to be less susceptible to IMD toxicity. SummaryThe role of ambient (21%) and physiological oxygen (2-5%) in the immortalization of fetal vs. adult human lung fibroblasts was examined. Growth in low oxygen and antioxidants extended the lifespan of both fetal and adult strains. As the ectopic expression of telomerase could immortalize adult lung fibroblasts cultured in ambient oxygen, the lifespan-shortening effects of 21% oxygen must have been largely limited to telomeres. By contrast, fetal lung fibroblasts could not be immortalized in ambient oxygen in spite of telomere elongation by telomerase, suggesting more widespread oxidative damage. The longterm culture requirements for the immortalization of WI-38 fetal lung fibroblasts included supplementation with N-( tert ) butyl hydroxylamine, dexamethasone, zinc and vitamin B12, in addition to growth in physiological oxygen. The mechanisms regulating telomere shortening remain controversial. The present results suggest that both endreplication and oxidative damage events contribute to telomere shortening in lung fibroblasts in vitro . These observations emphasize the need for better analytical techniques to distinguish whether the correlation of short telomeres with disease and mortality in humans reflects the consequences of increased proliferation, telomere shortening as a result of oxidative damage or some combination of these processes.
