Age-related development of a heterozygous phenotype in solitary neurons of the homozygous Brattleboro rat.

Abstract: A single-base deletion in the single-copy vasopressin gene is the cause of diabetes insipidus in the homozygous Brattleboro rat (di/di). It results in the synthesis of an altered vasopressin precursor of which the axonal transport is blocked. Paradoxically, a small number of solitary hypothalamic neurons displays all the immunoreactivities of the wildtype vasopressin precursor (i.e., vasopressin, neurophysin, and a glycopeptide). In the present paper we provide evidence that these neurons have undergone a swit… Show more

“…Because inhibition by the mechanism described here relies on the modification of the mutant protein by wild-type Ub, Ub ϩ1 should act in a dominant negative fashion. This feature is critical in view of the low efficiency of the misreading process (17)(18)(19). Van Leeuwen and coworkers (17) estimated that frame-shifted UbB transcripts comprise between 1% and 10% of the total UbB transcripts in the brains of AD patients.…”

“…Alternatively, cryosections were fixed in 4% paraformaldehyde. Sections were incubated with polyclonal anti-Otp (Lin et al 1999) diluted 1:500 in 50 mM Tris/0·5 M NaCl/0·5% Triton X-100 or a polyclonal antiserum raised against the glycopeptide of the vasopressin precursor hormone (C3 final; van Leeuwen et al 1989) diluted 1:5000, at 4 C overnight. Sections were then incubated with biotinylated goat anti-rabbit (Vector Laboratories) secondary antibody, diluted 1:1000 in Tris-buffered saline, and processed according to the ABC method (Vector Laboratories).…”

“…In the nuclear genome, inappropriate mutational inactivation of nuclear genes occurs with age (7), as well as inappropriate reactivation of silent genes (8)(9)(10). The accumulation of deleterious mutations in the mitochondrial genome has also been proposed to be important in aging (2)(3)(4)(5).…”
