Age-related epigenome-wide DNA methylation and hydroxymethylation in longitudinal mouse blood

Abstract: DNA methylation at cytosine-phosphate-guanine (CpG) dinucleotides changes as a function of age in humans and animal models, a process that may contribute to chronic disease development. Recent studies have investigated the role of an oxidized form of DNA methylation - 5-hydroxymethylcytosine (5hmC) - in the epigenome, but its contribution to age-related DNA methylation remains unclear. We tested the hypothesis that 5hmC changes with age, but in a direction opposite to 5-methylcytosine (5mC), potentially playin… Show more

“…Therefore, it is understandable that DNA methylation level was higher in TYO sh. Similar result also reported in mouse, which blood showed a general pattern of epigenome-wide hypermethylation with age [43]. The DNA hypermethylation in TYO sh may be involved in the epigenetic reprogramming during development, which is important in regulate gene expression [44,45].…”

“…Only one study has examined perinatal BPA exposure effects on liver histone modifications, and the effect of these modifications on gene expression and liver function were not reported 44 . In contrast to the little that is known regarding the impact of epigenome:environment interactions on histone modifications, aging-associated changes in DNA methylation (DNAm-aging) have become well-established since the original studies of Horvath 45 , and reflect the impact of both environmental exposures and pathophysiological processes 41,[46][47][48][49][50][51] . Previous studies have assessed epigenetic signatures in rodent and human liver during normal aging by assessing DNAm-aging [52][53][54][55] .…”

“…Rates of 5hmC were then natural log (ln) transformed to achieve a normal distribution for the linear regression analysis, which was used to examine the association between categorized BPA exposure and 5hmC. Age, smoking, alcohol consumption, and history of disease, which had been reported to be associated with 5hmC [39,40,41], were adjusted for as potential confounders.…”
