Aggregated Tau activates NLRP3–ASC inflammasome exacerbating exogenously seeded and non-exogenously seeded Tau pathology in vivo

Abstract: Brains of Alzheimer’s disease patients are characterized by the presence of amyloid plaques and neurofibrillary tangles, both invariably associated with neuroinflammation. A crucial role for NLRP3–ASC inflammasome [NACHT, LRR and PYD domains-containing protein 3 (NLRP3)–Apoptosis-associated speck-like protein containing a CARD (ASC)] in amyloid-beta (Aβ)-induced microgliosis and Aβ pathology has been unequivocally identified. Aβ aggregates activate NLRP3–ASC inflammasome (Halle et al. in Nat Immunol 9:857–865,… Show more

“…Meisoindigo reduced brain water content and significantly downregulated the expression of AQP4 compared with the vehicle group (P < 0.05) (Figure 2). MCC950, a highly potent and selective inhibitor of the NLRP3 inflammasome, in vitro and in vivo (Ren et al, 2018;Stancu et al, 2019), had effects similar to those of meisoindigo.…”

“…Phagocytosed tau can be released in exosomes to promote tau propagation in adjacent cells [78]. Tau aggregates like prions and the aggregated tau activates the nod-like receptors P3-apoptosis-associated speck-like protein containing caspase activation and recruitment domains (CARD) (ASC) inflammasome, one of the sensors important for activation of innate immune response [79]. Early microglial activation and central and peripheral inflammation were detected in patients with dementia with LBs [80].…”

“…Bot deletion of tau and hyperphosphorylation of tau impair insulin signaling (Marciniak et al, 2017;Rodriguez-Rodriguez et al, 2017). Tau can also induce inflammasome activation in the brain, which may further contribute to increased reactive oxygen species (ROS) and impaired insulin signaling (Ising et al, 2019;Stancu et al, 2019). Thus, it appears that the Aβ and tau proteins likely have a shared role in causing dysregulated insulin signaling in the brain.…”
