Aging-Associated Cognitive Decline is Reversed by D-Serine Supplementation

Abstract: Brain aging is a natural process that involves structural and functional changes that lead to cognitive decline, even in healthy subjects. This detriment has been associated with NMDA receptor (NMDAR) hypofunction because of a reduction in the brain levels of D-serine, the endogenous NMDAR co-agonist. However, it is not clear whether D-serine supplementation could be used as an intervention to reduce or reverse age-related brain alterations. In the present work, we aimed to analyze the D-serine effect on aging… Show more

“…In addition, dynamic interactions between the OFC and hippocampus for reversal learning have been noted, with local field potentials of the OFC and hippocampus exhibiting coherence in theta rhythm in a performancedependent manner [78,79] and contralateral lesions of the OFC and ventral hippocampus in mice subtly, though significantly, impairing spatial reversal learning [78]. Systemic D-serine treatment has shown beneficial effects on spatial reversal learning [69,[80][81][82][83]. However, for this study, upregulation of SR in the mPFC did not affect reversal.…”

“…(d) The NMDA receptor surface expression and its functions also alter with age. NMDA receptor hypofunction, as well as a decline of calcium entry via NMDA receptors were reported in elderly (Foster, 2007;Nava-Gómez et al, 2022).…”

“…Regulating D-serine levels through SR can be a novel drug target for the treatment of neurotoxic disorders like stroke and traumatic brain injury or neurodegenerative disorders including amyotrophic lateral sclerosis (ALS) and late-stage Alzheimer's disease (AD) with elevated D-serine [8][9][10][11][12][13][14]. At the same time, treatment with an NMDA receptor agonist, D-serine alone, or D-serine combined with a glycine reuptake inhibitor has been demonstrated to improve the psychotic symptoms and cognition in patients with schizophrenia, depression, early-stage AD, and agingassociated cognitive decline [7,[15][16][17][18]. In a double-blind crossover clinical trial, D-serine treatment could alleviate symptoms of schizophrenia and depression [19].…”
