AgNORs Predictive Value of Prognosis in Non-Hodgkin's Lymphoma: Comparison with Flow Cytometric Cell Cycle Analysis

Abstract: Paraffin-embedded histopathologic specimens, taken before the commencement of therapy from 14 low-grade and 21 high-grade malignant lymphoma patients, and 9 normal lymph nodes were utilized to analyze six cell DNA-related parameters. The flow cytometry technique was used to determine cell-cycle G0/G1, S and G2/M phases, and silver staining to enumerate nuclear organized regions (AgNORs); nucleus surface area was determined by an image-analyzing system. The six parameters and natural logarithm of cell proportio… Show more

“…36 In lymphoid malignancies, relatively few studies pertain to DNA ploidy and morphometrically ascertained AgNOR count: there are only some instances of the prognostic significance of DNA analysis [37][38][39] and the association of high AgNOR numbers with increased proliferative activity and poor clinical outcome in high-grade lesions. 40 Moreover, in low-grade lymphomas the AgNOR maximum diameter ranged from 0·7 to 1·7 m (mean 1·11 m) and area ranged from 0·48 to 1·99 m 2 (mean 1·11 m 2 ) while in high-grade lymphomas the maximum diameter ranged from 0·33 to 0·51 m (mean 0·41 m) and area ranged from 0·082 to 0·19 m 2 (mean 0·13 m 2 ). This demonstrated that an inverse relationship did exist between AgNOR numbers and their sizes.…”
