Agomelatine Improves Sleep in a Patient with Fatal Familial Insomnia

Abstract: A young patient with FFI was started on agomelatine 25 mg to medicate nocturnal insomnia. Under this treatment sleep efficiency was improved, slow wave sleep was high and awakenings during sleep period time were far less than before. “…More importantly, our REM, NREM, SE, and TST results were consistent with previous investigations that undertook 8 weeks of escitalopram treatment in depression subjects 36 . The improvement of sleep quality, including TST, SE, and REM, was also observed in the melatonin receptor agonist and serotonergic receptor antagonist, agomelatine 37,38 …”

Namely, SNP might regulate rhythm by advancing the secretion phase of melatonin.…”

“…Before its marketing AGM had demonstrated antidepressive eff ects in several animal models of depression and furthermore featured a potential to restore normal circadian rhythms in animal models of a disrupted circadian system [ 6 ] . In line with this there is clinical evidence that AGM improves sleep in familial insomnia [ 21 ] . According to published results of clinical trials AGM had proven superior effi cacy in comparison to placebo [ 22 -26 ] and equivalent effi cacy compared to standard antidepressive agents in the treatment of the acute phase Introduction ▼ Agomelatine (AGM), an acetamide naphthalene analogue of melatonin, was approved by the European Medicines Agency (EMA) for the treatment of major depressive disorder in adults in February 2009 [ 1 ] .…”
