Agrin-Matrix Metalloproteinase-12 axis confers a mechanically competent microenvironment in skin wound healing

Abstract: An orchestrated wound healing program drives skin repair via collective epidermal cell proliferation and migration. However, the molecular determinants of the tissue microenvironment supporting wound healing remain poorly understood. Herein we discover that proteoglycan Agrin is enriched within the early wound-microenvironment and is indispensable for efficient healing. Agrin enhances the mechanoperception of keratinocytes by augmenting their stiffness, traction stress and fluidic velocity fields in retaliatio… Show more

“…The emergence of regenerated hair follicles and dermal papilla in the center of the wound area (Fig. 4C–l ) may suggest that ADSCs embedded in the DA-MeHA hydrogel significantly accelerated skin remodeling [ 16 ]. ADSCs encapsulated by the DA-MeHA hydrogel may differentiate into primary mesenchymal cells, accelerate the regeneration of surrounding hair follicles, and simultaneously induce new blood vessel generation.…”

“…Cells probe the stiffness of their niche using mechanotransduction to interact with the ECM and surrounding cells, sense mechanical perturbations, and transmit environmental pressure to modulate their behaviors [ 15 ]. In addition to functions of cell scaffolding, the ECM serves as a dynamic communicative plane to its circumambient cells in the tissue to maintain tissue integrity [ 16 ]. Hydrogels are characterized by unique hydrated networks and physicochemical properties and have been widely used as scaffolds for cell therapies based on their ability to mimic the ECM [ 13 , 17 ].…”

“…Apart from MMP1 and MMP3 , MMP12 and MMP9 were upregulated. MMP-12 appears to play a key role in cytoskeletal rearrangements in migrating keratinocytes [ 35 ]. MMP-9 protein increases during suction blister wound healing [ 7 , 11 ].…”

“…MMP-7 is responsible for stimulating the process of re-epithelialization ( Letra et al, 2013 ). MMP-12 is induced by Agrin and mediates collective migration due to enhanced local inflammation ( Aristorena et al, 2019 ; Chakraborty et al, 2021 ). Although chronic cutaneous wounds in humans have not been extensively studied, these MMPs play a key part in wound healing because of their ability to influence variety of cellular processes and activate other MMPs.…”
