AgroLux: bioluminescent Agrobacterium to improve molecular pharming and study plant immunity

Abstract: SUMMARY Agroinfiltration in Nicotiana benthamiana is widely used to transiently express heterologous proteins in plants. However, the state of Agrobacterium itself is not well studied in agroinfiltrated tissues, despite frequent studies of immunity genes conducted through agroinfiltration. Here, we generated a bioluminescent strain of Agrobacterium tumefaciens GV3101 to monitor the luminescence of Agrobacterium during agroinfiltration. By integrating a single copy of the lux operon into the genome, we generate… Show more

“…Once the Agrobacteria are inside the leaf, DNA encoding the cargo is transferred into leaf cells leading to transient expression of the cargo. Historically, agroinfiltration has been valuable in phenotyping the plant immune system (Ma et al ., 2012; Bücherl et al ., 2017; Jutras et al ., 2021).…”

“…In some cases, the image processing software ImageJ is used to obtain numerical data from images obtained with ultra-sensitive CCD cameras (21,28). The throughput of the former approach is low and the ImageJ macros used are often not well documented (21,24). Hence, reproducibility is poor and/or still requires manual preparation of the individual sample images (8).…”
