Air pollution and diabetes association: Modification by type 2 diabetes genetic risk score

Abstract: Exposure to ambient air pollution (AP) exposure has been linked to type 2 diabetes (T2D) risk. Evidence on the impact of T2D genetic variants on AP susceptibility is lacking. Compared to single variants, joint genetic variants contribute substantially to disease risk. We investigated the modification of AP and diabetes association by a genetic risk score (GRS) covering 63 T2D genes in 1524 first follow-up participants of the Swiss cohort study on air pollution and lung and heart diseases in adults. Genome-wide… Show more

“…Although there are at least seven common variants on MTNR1B influencing glucose metabolism through a common pathway [ 21 ], sleep and circadian studies have only focused on rs10830963 variant [ 24 ]. In essence, following the approach of using scores involving multiple variants for improved prediction of genetic risks [ 25 , 26 ], an MTNR1B genetic risk score (MGRS) based on variants that are not in strong linkage disequilibrium (LD), may better capture individuals at high-risk of melatonin profile dysregulation than single variants.…”

“…Swiss TPH has a track record in IDs and NCDs research, which forms the basis for the scientific objectives of this project. Swiss TPH is running the only Swiss-wide chronic disease cohort and biobank (Swiss Cohort Study on Air Pollution and Lung and Heart Diseases in Adults), participates at the forefront of exposome research [ 49 , 71 , 74 ], and its coordinators have vast experience in biobanking (Swiss Biobanking Platform). The Université Félix Houphouët-Boigny is the largest public university in Côte d’Ivoire with several laboratories covering ecology, parasitology, and zoology.…”
