Alarmins, COVID-19 and comorbidities

Abstract: The coronavirus SARS-CoV-2, the aetiological agent of COVID-19 disease, is representing a worldwide threat for the medical community and the society at large so that it is being defined as “the twenty-first-century disease”. Often associated with a severe cytokine storm, leading to more severe cases, it is mandatory to block such occurrence early in the disease course, to prevent the patients from having more severe, sometimes fatal, outcomes. In this framework, early detection of “danger signals”, possibly re… Show more

“…The use of such cytokine-controlling antibodies can significantly improve the efficacy of neutralizing antibody therapy. Alarmins, such as HMGB1, IL-31, IL-33, and S100, have been shown in some studies on the mechanism of cytokines to aggravate cytokine storm and disease [ 87 ]. Alfarouk et al described the specific mechanism of cytokine storms from the perspective of mitochondria: mitochondria are the focal point of cytokine storms.…”

“…Recent studies have pointed to the significance of early detection of danger signals for the classification of COVID-19 patients as being at high risk of mortality. Alarmins are possible danger signals associated with COVID-19 and comorbidities [63], and S100A8/A9, high mobility group box 1 (HMGB1), and histones are considered potential therapeutic targets [64][65][66]. The elevation of S100A8/A9 and HMGB1 by Ang-II suggests the involvement of alarmins in unbalanced RAS [67,68], and the ACE2/Ang-(1-7)/MasR axis could suppress HMGB1 signaling [69].…”
