Alleles ofafd1dissect REC8 functions during meiotic prophase I

Abstract: REC8 is a master regulator of chromatin structure and function during meiosis. Here, we dissected the functions of absence of first division (afd1), a maize rec8/α-kleisin homolog, using a unique afd1 allelic series. The first observable defect in afd1 mutants is the inability to make a leptotene chromosome. AFD1 protein is required for elongation of axial elements but not for their initial recruitment, thus showing that AFD1 acts downstream of ASY1/HOP1. AFD1 is associated with the axial and later the lateral… Show more

“…In budding yeast, the meiosis-specific cohesion component REC8 binds preferentially to AT-rich regions (axis association sites) during the development of axis structures to produce co-oriented linear arrays of loops (Zickler and Kleckner, 1999;Blat et al, 2002). In Arabidopsis and maize (Zea mays), homologs of REC8 are required for homologous pairing and recombination, largely by influencing the chromatin structure (Bhatt et al, 1999;Cai et al, 2003;Chelysheva et al, 2005;Golubovskaya et al, 2006). Rice REC8 was recently identified, and its depletion resulted in a lack of homologous pairing and failure to load key rice meiotic proteins, including PAIR2, PAIR3, MER3, and ZEP1, onto the chromosomes (Wang et al, 2009Shao et al, 2011).…”

“…For example, in maize, impairments in AFD1 result in defective telomere bouquets, indicating that the elongation of AEs on chromosomes is required for bouquet formation [10]. In rice, installation of PAIR2 depends on the normal formation of AE.…”

“…Cohesin proteins, which are thought to be involved in this process, have been extensively studied in yeast and animals [4][5][6]. In plants, some cohesin proteins have been characterized for their influence in meiotic pairing, especially the REC8 homologs: SYN1/DIF1 in Arabidopsis [7][8][9], AFD1 in maize [10] and OsRAD21-4 in rice [11]. It is becoming evident that the cohesin complex provides the basis for AE assembly and is implicated in various meiotic events [4,12].…”

“…The mutation of Rec8 of yeast disrupts cohesion at both chromosome arms and centromeres, resulting in reduced recombination, alterations in synaptonemal complex formation, and premature separation of sister chromatids at meiosis I (Klein et al, 1999;Watanabe and Nurse, 1999;Nasmyth, 2001;Kitajima et al, 2003). In recent years, homologs of yeast REC8 have been identified in mice (MmRec8; Bannister et al, 2004), humans (HsRec8; Parisi et al, 1999), Caenorhabditis elegans (CeRec8; Pasierbek et al, 2001), Arabidopsis (Arabidopsis thaliana; SYN1; Bai et al, 1999), and maize (Zea mays; AFD1; Golubovskaya et al, 2006). Depletion of this protein leads to defects in meiosis.…”

“…By contrast, SYN1 is necessary not only to maintain centromere cohesion at anaphase I but also for the monopolar orientation of kinetochores during the first meiotic division in Arabidopsis (Chelysheva et al, 2005). In maize, AFD1/REC8 controls homologous pairing through its role in axial element elongation and the subsequent distribution of the recombination machinery (Golubovskaya et al, 2006). Rice (Oryza sativa) is a model organism for monocots that represents another excellent system for studying the molecular mechanism of meiosis in high plants.…”
