Alpha/Beta-Hydrolase Domain-Containing 6: Signaling and Function in the Central Nervous System

Abstract: Endocannabinoid (eCB) signaling plays an important role in the central nervous system (CNS). α/β-Hydrolase domain-containing 6 (ABHD6) is a transmembrane serine hydrolase that hydrolyzes monoacylglycerol (MAG) lipids such as endocannabinoid 2-arachidonoyl glycerol (2-AG). ABHD6 participates in neurotransmission, inflammation, brain energy metabolism, tumorigenesis and other biological processes and is a potential therapeutic target for various neurological diseases, such as traumatic brain injury (TBI), multip… Show more

“…CPT1C is a member of the carnitine palmitoyltransferase 1 family and is involved in the regulation of physiological functions such as energy metabolism and feeding ( 38 ). CPT1C can have profound effects on brain physiology and total fatty acid profiles, which can be modulated by nutrients in the diet ( 39 ).…”

“…This does not, however, rule out the possibility that the hiPSC-derived cultures can produce other eCBs, perhaps with different stimuli. Furthermore, the carbachol-evoked GRAB eCB2.0 response continued to recover in the presence of JZL 184 despite being significantly slowed, suggesting that other degradative enzymes such as Alpha/Beta-Hydrolase Domain-Containing 6 (Zhang et al, 2021) might be present in hiPSCderived neuronal cultures. There are multiple ways to evoke eCB synthesis, with different levels of Ca 2+ dependence.…”

“…5). ABHD6, a hydrolase of monoacylglycerols, regulates levels of the most abundant endocannabinoid, 2-araquidonoylglycerol, which is a modulator of neurotransmitter release through its binding to the endocannabinoid receptor CB1 at the neuron presynaptic membrane [54]. Importantly, CB1 has also been found in the external mitochondrial membrane, where it directly controls mitochondrial respiration and energy production [55].…”
