Alpha‐Lipoic Acid Protects against Indomethacin‐Induced Gastric Oxidative Toxicity by Modulating Antioxidant System

Abstract: Gastroprotective effects of α-lipoic acid (ALA) against oxidative gastric damage induced by indomethacin (IND) have been investigated. All doses (50, 75, 100, 150, 200, and 300 mg/kg body weight) of ALA reduced the ulcer index with 88.2% to 96.1% inhibition ratio. In biochemical analyses of stomach tissues, ALA administration decreased the level of lipid peroxidation (LPO) and activities of myeloperoxidase (MPO) and catalase (CAT) in gastric tissues, which were increased after IND application. ALA also increas… Show more

“…Lipoic acid protects against the effects of Bisphenol A or Bi-n-butyl phthalate on testicular mitochondrial toxicity [210,211]. In various other models it also protects against the toxic effects of acetaminophen [212], acrolein [213], cyclosporine [214], indomethacin [215], paraquat [216] and rotenone [217] as well as cypermethrin [218], dimethoate, glyphosate and zineb [219],chrysene [220] lindane [221] and Tetrachlorodibenzodioxin [222]. Lipoic acid and other antioxidants have also been used in the clinical management and prevention of heavy metal intoxication [223].…”

“…In this study, the activities of GSH-dependent enzymes, viz., GPx and GST, were significantly decreased in ISOtreated rats relative to the control. The sharp fall in GPx and GST activities may be linked to increased oxidative stress causing a decrease in GSH levels and ultimately affecting the activities of GPx and GST [33,34]. Furthermore, the ISO-induced myocardial injury was associated with significantly lowered levels of other antioxidant enzymes (CAT and SOD).…”

“…Briefly, vitamin B12 was found to be inadequate in avoiding the structural changes after trauma such as edema, myelin sheath degeneration, polyaxonal Schwann cells, e-dense depositions in Schwann cell cytoplasm and abnormal While the antioxidant propery of A-LA is well known as reported in other previous studies report (13,16), the presented study basically aimed to demonstrate the effects of long term use of A-LA and vitamin B12 histomorphologically and structurally.…”
