Alpha-skeletal muscle actin mRNA's accumulate in hypertrophied adult rat hearts.

Abstract: Cardiac hypertrophy due to a chronic hemodynamic overload is accompanied by isoformic changes of two proteins of the thick filament of the sarcomere, myosin, and creatine phosphokinase. We have looked for isoactin changes, using deoxyribonucleic acid probes complementary to alpha-skeletal and alpha-cardiac actin messenger ribonucleic acids. Three groups of rats were studied at various days after application of a pressure overload (2-4 days, n = 13, 8-15 days, n = 5, and 30-40 days, n = 7) and were compared to … Show more

“…The increases in the expression levels of: (a) calreticulin, which has also been reported to increase in pressure overload hypertrophy (42) In the calsequestrin overexpressing heart as well as a variety of other experimental models, cardiac hypertrophy was associated with increased expression of fetal genes (23,(43)(44)(45). Interestingly, the up-regulation of ␣-skeletal actin and ANF mRNA was reversed by phospholamban ablation.…”

“…Compensatory expression of skeletal ␣-actin is also seen in adult BALB͞c mice, which have a mutant cardiac ␣-actin locus and decreased cardiac ␣-actin levels (2). Skeletal and vascular smooth muscle ␣-actin mRNAs accumulate in the adult rat heart, the former transiently, following hypertrophy induced by pressure overload (7,39,40). While the pattern of compensation in the newborn ϩ͞Ϫ and Ϫ͞Ϫ hearts is in some ways reminescent of the expression seen in the hypertrophied hearts, there are no morphological changes in the newborn ϩ͞Ϫ hearts that would suggest hypertrophy (not shown).…”

“…Similarly, there is a preponderance of cardiac ␣-actin mRNA in the heart of the adult chicken (4), mouse (2,5), and rat (6)(7)(8). However, the adult human heart has more skeletal than cardiac ␣-actin mRNA (9).…”

“…hypertension), often leads to irreversible cardiomyopathy and heart failure. The pathological hypertrophic response is characterized by the transcriptional activation of fetal genes encoding contractile proteins such as ␤-MHC and skeletal ␣-actin (8,9). The synthesis of atrial natriuretic factor (ANF) is also induced, presumably mimicking the cardiac response to pressure overload (10).…”
