Alteration of the L‐type calcium current in guinea‐pig single ventricular myocytes by heptaminol hydrochloride

Abstract: 1 The effects of heptaminol on calcium current amplitude and characteristics were studied in single ventricular myocytes of guinea-pig by use of the whole cell configuration of the patch clamp technique. 2 A concentration-dependent decrease in 'Ca amplitude was observed. At heptaminol concentration as low as 10-6 M, this effect was observed in only two cells (n = 6). At 10-5 M the reduction of ICa was of 30± 15% (n =11). 3 The current recovery from inactivation at -40 mV holding potential (HP) seemed less sens… Show more

“…Among the top 10 compounds, trichostatin A (Figure 4B) and heptaminol (Figure 4C) had the highest binding affinity to CDK1, which was mutually confirmed by previous studies [48]. Furthermore, heptaminol has been proven to treat cardiovascular system problems such as anti-hypotensive action, right heart failure, and cardiogenic edema [49,50], therefore it was selected as the key drug.…”

“…The chemical structures of these compounds were also downloaded (Figure 4A) from the ZINC database (http://zinc15.docking.org). Among these small compounds, previous studies have proven that heptaminol [44,45] and tranexamic acid [46] can be used in cardiovascular diseases, while the others have the effects of anti-tumor or antiinflammatory, etc.…”

“…It has been used to treat low blood pressure and orthostatic hypotension ( Boismare and Boquet, 1975 ), with reported ‘unexpected’ adverse effects ( Pathak et al ., 2005 ). Its actions may involve NA release by action at the transporter ( Grobecker and Grobecker, 1976 ; Delicado et al ., 1990 ), but it may also have effects on Ca2+ channels to increase intracellular calcium ( Peineau et al ., 1992 ). See also Isometheptene.…”

“…Pindolol was chosen over carvedilol for current study due to its lack of α-adrenoreceptor blocking activity. Heptaminol, on the other hand, is a vasodilator and a general antagonist to catecholamine release and uptake that also raises intracellular free calcium level [12, 13]. Lastly verapamil is an L-type calcium channel blocker.…”
