Alterations in the profile of blood neutrophil membrane receptors caused by in vivo adrenocorticotrophic hormone actions

Abstract: Elevated levels of adrenocorticotrophic hormone (ACTH) mobilize granulocytes from bone marrow into the blood, although these neutrophils are refractory to a full migratory response into inflamed tissues. Here, we investigated the dependence of glucocorticoid receptor activation and glucocorticoid-regulated protein annexin A1 (ANXA1) on ACTH-induced neutrophilia and the phenotype of blood neutrophil after ACTH injection, focusing on adhesion molecule expressions and locomotion properties. ACTH injection (5 μg i… Show more

“…We have previously shown that AnxA1 −/− mice present neutrophilia (Dalli et al, ; Machado et al, ), and show here that the circulating pool of neutrophils in AnxA1 −/− mice is characterized by higher and reduced numbers of CXCR4 + and CXCR2 + neutrophils, respectively. This profile of cells in the circulation was reversed if AnxA1 −/− mice were treated for 4 days with the peptide, Ac2‐26 (Fig.…”

“…182,183 In contrast, the injection of an adrenocorticotrophic hormone induced upregulation of CXCR4 on murine neutrophils in vivo. 184 Multiple lines of evidence suggest a pivotal role for the CXCL12-CXCR4 axis in the regulation of neutrophil homeostasis (Figs. 1 and 4).…”

“…We must admit that not all patients showed reduced CXCR2 levels. However, it has been shown that in some circumstances chemokine receptors, although being upregulated, transmit migratory responses less efficiently ( 17 19 ).…”
