aThe prevalence of obesity and other obesity-related diseases is increasing worldwide. Obesity is a disease characterized by increased body weight, or a condition resulting from excessive accumulation of body fat. Due to increased body fat deposits, obesity has also been associated with increased mortality resulting from higher incidence rates of hypertension, diabetes, or various types of cancer, such as breast, colorectal, cervical and prostate cancer. Physiological changes associated with obesity are likely to result in altered drug biotransformation. The main enzymes enabling the oxidative biotransformation of most drugs are cytochromes P450 (CYPs). The review summarizes how pathophysiological factors, especially obesity, affect properties (e.g. enzyme activity, protein expression, gene expression) of CYP enzymes in various experimental models of human obesity. Results reported by various authors suggest that obesity is associated with a decrease of CYP activities (except for the CYP2C and CYP2E1 enzymes). The only exception is mouse obesity induced by monosodium glutamate (administered to newborn mice) as it usually leads to increased CYP expression. Selecting an animal model that is as close as possible to the properties of human obesity is of paramount importance.

Background: Hepatic cytochrome P450 (CYP) isoforms, CYP1A2, is one of important enzymes for many drugs metabolism. Studies have confirmed that sustained hypoxia can influence the expression of hepatic CYP, including CYP1A2. The impact of chronic intermittent hypoxia (CIH), a marked characteristic of sleep apnea, on CYP1A2 remains unclear. The aim of the present study was to evaluate the effect of CIH on the expression of hepatic CYP1A2 in a mouse model with sleep apnea.Methods: Twenty four old male (6-8 weeks) C57BL/6J mice (n=12 in each group) were randomly assigned to either normoxia group or CIH group. Mice in CIH group underwent 12 weeks intermittent hypoxia exposure. The different gene expression of hepatic CYP1A2 between two groups was analyzed by quantity real-time polymerase chain reaction. The protein levels of hepatic CYP1A2 in each group were observed by using western blotting and immunohistochemistry.Results: After 12 weeks of exposure to intermittent hypoxia, the expression of hepatic CYP1A2, at the mRNA and protein levels was decreased more significantly in the CIH group than the normoxia group (P<0.01).Conclusions: CIH contributes to inhibiting the expression of hepatic CYP1A2. This implies that the dosage of drugs metabolized by CYP1A2, should be adjusted in patients with sleep apnea.

Key words: monosodium glutamate-induced obesity, catechins, oxidative stress, polyphenon 60Green tea and green tea extracts (GTE) are often incorporated into diet intended to weight reduction, although the information about their effi cacy in obese individuals is insuffi cient. The present study was designed to follow up the effect of defi ned and standardized GTE in mice with obesity induced by monosodium L-glutamate. Obese mice were fed with GTE-supplemented diet in three dosage regimens: 28-day and 3-day intake of 1 g GTE in 1 kg of diet and 28-day intake of 0.1 g GTE in 1 kg of diet. The information on body weight, food intake, oxidation stress parameters in blood and antioxidant enzymes activity in liver and small intestine was obtained. High doses of GTE decreased the specifi c activities of glutathione reductase and catalase and increased concentrations of malondialdehyde in blood. Specifi c activities of antioxidant enzymes in the liver and small intestine were not altered after GTE treatment except the decrease of NAD(P)H:quinone oxidoreductase activity. Our results showed that GTE did not affect average body weight and did not markedly improve antioxidant status in glutamate-induced obese mice.
