Alternative splicing isoform of tau protein kinase I/glycogen synthase kinase 3β

Abstract: Glycogen synthase kinase 3 (GSK3) plays important roles in Wnt and insulin signaling, cell fate determination, and Alzheimer-like tau phosphorylation. We discovered an isoform of tau protein kinase I (TPKI) / GSK3b with a 13 amino acid insert in the catalytic domain owing to alternative splicing. The alternative transcripts were found in the brains of the mouse, rat and human, with highly conserved sequences. The variant protein, named TPKI2 / GSK3b2, was abundant in the brain. Immunohistochemistry indicated d… Show more

“…Moreover, the addition of 50 M TWS119 to the lysates during affinity chromatography effectively blocked binding of the 47-and 49-kDa proteins to the affinity matrices, further demonstrating specificity for TWS119. These two bands were identified to be GSK-3␤ by LC͞MS, and possibly represent two different splicing isoforms (18) or posttranslationally modified products. More specifically, six individual tryptic-digest peptide fragments of GSK-3␤ were identified with estimated false positive rates of Ͻ1% (19).…”

“…The two isoforms have high amino acid sequence identity (97%) in the catalytical domain, but are less conserved otherwise. GSK3 has two splicing isoforms, one containing a 13 aa insertion (GSK3 2) [100]. Although mutations in GSK3 are usually not associated with cancers, downregulation of GSK3 has been observed in hepatocellular carcinoma, squamous cell carcinoma and prostate cancer [101][102][103].…”

“…Both enzymes retain activity when their non-catalytic N-terminal extensions are deleted, whereas truncation mutants of their disparate non-catalytic C-terminal sequences display reduced activity, in part due to improper folding (Buescher and Phiel, 2010). There is a splice variant of GSK-3 (GSK-3 2), which has a 13 amino acid insert in the kinase domain (Mukai et al, 2002;Schaffer et al, 2003). The GSK-3 2 variant is less abundant than the GSK-3 1 variant.…”
