AMP-Activated Protein Kinase as a Target for Preconditioning in Transplantation Medicine

Abstract: Graft quality before transplantation is a major factor influencing chronic rejection. Organ preservation and ischemia/reperfusion play an important role in the induction of organ injury. Although both suppression of metabolism by hypothermic preservation and preconditioning before ischemia limit injury, understanding the biochemical signaling pathways will allow us to optimize graft preservation further. Adenosine monophosphate-activated protein kinase (AMPK) is an important enzyme sensing cellular energy bala… Show more

“…There was also a solution effect on cortical microcirculation after unclamping reflected by higher laser Doppler values in the MPS groups than in the UW groups. Further studies should evaluate the role of AMPKa in the solution effect on cortical microcirculation because AMPKa has been reported to play a role in the protective mechanisms of preservation solutions (45) and in the ischemic preconditioning of liver grafts (46).…”

“…Similar results were observed when CE and IGL1 were compared. Moreover, in partial grafts, IGL1 provided better survival than the other solutions because it increased the mediators that promote liver regeneration such as AMP-activated protein kinase (AMPK) (Bouma et al 2010). Furthermore, UW and IGL1 solutions enriched with trophic factors, such as epidermal growth factor and insulin-like growth factor-1, are observed to enhance the resistance of steatotic livers …”
