Amygdala Kindling Modifies Extracellular Opioid Peptide Content in Rat Hippocampus Measured by Microdialysis

Abstract: Opioid peptide release in the hippocampus was shown to be increased immediately following amygdala kindling stimulation in freely moving rats using microdialysis combined with a universal opioid peptide radioimmunoassay (RIA). Extracellular opioid peptide levels were elevated (55% above basal levels) within the first 10 min after electrical stimulation‐induced partial seizures in previously nonkindled animals. Fully kindled rats showed lower extracellular opioid peptide levels (40% reduction) during the interi… Show more

“…In turn, after kindling establishment, the increased degrading activity by the combined action of NEP and APN in hippocampus may explain the diminished extracellular opioid peptide levels detected during the interictal period, compared to those measured 10 min after stimulation or, in sham animals (Rocha et al 1997). Although there is little evidence regarding the involvement of mammillary nuclei in epileptogenesis (Mirski and Ferrendelli 1987), there is a strong and regulated expression of NEP; these cells project to limbic and extrapyramidal centers enriched with NEP protein and, with some of its substrates as substance P, bradykinin, neurotensin, and enkephalins (Gaudoux et al 1993).…”

“…Partial amygdaloid kindling increases the expression of proenkephalin and prosomatostatin (Burazin and Gundlach 1996;Rosen et al 1992;Shinoda et al 1991), the release of dynorphin A and leu-enkephalin in hippocampus, and met-enkephalin in amygdala immediately after stimulation (Rocha et al 1997). Once kindling is established, prodynorphin and prosomatostatin messenger RNA levels as well as opioids and somatostatin release are increased in hippocampus immediately after stimulation (Burazin and Gundlach 1996;Marti et al 2000;Rocha et al 1997;Romualdi et al 1999) but, after 24 h proenkephalin mRNA levels are increased and those of prodynorphin decreased in hippocampus and olfactory tubercle (Shinoda et al 1991). The stage-dependent regulation of NEP and APN expression may thus occur due to the particular balance of opioids expressed at each stage or to the extent (amount and time) of the provoked substrate release.…”
