An Analysis of the Optimal Conditions for Perfusing an Isolated Rabbit Kidney with Homologous Blood

Abstract: Isolated rabbit kidneys were perfused in a closed-circuit system with a number of media such a Krebs-Henseleit bicarbonate buffer with albumin, homologous platelet-free plasma, serum and whole blood. All of these media were unsatisfactory. In contrast, fresh blood deprived of platelets, leukocytes, floating lipoproteins, microaggregates and micro-clots did not cause vasoconstriction of the isolated kidney. Renal blood flow, glomerular filtration rate, urinary flow and glucose, cations and renal water reabsorpt… Show more

“…The isolated organ perfusion model used here has a number of advantages. The porcine kidney is recognized to have anatomical and physiological characteristics that closely resemble the human situation and as such the present model is likely to provide data that are more representative than those from small animal models12, 13. The pig organs used were taken from animals weighing 60–70 kg and this provided kidneys of similar size to adult human kidneys.…”

“…In order to better understand these events, we turned to blood perfusion as a suitable technique to measure the function of the isolated, perfused kidney [21]. A computer-controlled apparatus has been previously developed to assess in vitro renal function under normothermic conditions using blood as a perfusate [l].…”

“…All kidneys experienced transient reductions in renal blood flow, and the severity of the vascular response appeared to determine the ability of the kidney to produce urine and function within the period of evaluation. However, the pathophysiologic basis of this transient hypoxia (vascular crisis) was not elucidated in either the ex vivo or the transplant model [ l l , 14, 15, 161. In order to better understand these events, we turned to blood perfusion as a suitable technique to measure the function of the isolated, perfused kidney [21]. A computer-controlled apparatus has been previously developed to assess in vitro renal function under normothermic conditions using blood as a perfusate [l].…”
