An atherogenic diet rapidly induces VCAM-1, a cytokine-regulatable mononuclear leukocyte adhesion molecule, in rabbit aortic endothelium.

Abstract: Accumulation of monocyte-derived foam cells in focal areas of the arterial intima is a key step in early atherogenesis. We investigated the expression of vascular cell adhesion molecule-1 (VCAM-1), a mononuclear leukocyte adhesion molecule, in the arterial endothelium during the early phases of diet-induced atherogenesis in rabbits in vivo and the regulation of VCAM-1 expression by cytokines in rabbit aortic organoid cultures in vitro. Rabbits were fed either an atherogenic diet (containing 0.3% cholesterol, 9… Show more

“…This occurs in part through the induction of early atherogenic inflammatory responses, including the expression of adhesion molecules like VCAM1 by endothelial cells (ECs) (3,4). Consistent with this, increased dietary cholesterol rapidly induces atherosclerosis in animal models, with changes in VCAM1 expression seen within 2 weeks (3).…”

“…This is likely due in part to both paracrine and autocrine mechanisms by which the cytokines IL-1l3 and TNFa, derived from both inflammatory and endothelial cells, induce the cellular synthesis ofreactive oxygen species ( 19,20). It is in this oxidative milieu that endothelial VCAM-I expression and consequent monocyte accumulation is observed in the early atherogenic lesion (3,21 ). These observations suggest that the activation of VCAM-1 gene expression might be distinguished by its sensitivity to modulation by oxidation-mediated signals.…”

“…Receptors such as vascular cell adhesion molecule-1 and intercellular adhesion molecule-1 are up-regulated on the endothelium in response to noxious stimuli (8). These molecules couple with the integrins expressed on rolling leukocytes and adhere these leukocytes to the endothelium (9).…”
