An Engineered Pathway for the Formation of Protein Disulfide Bonds

Abstract: We have engineered a pathway for the formation of disulfide bonds. By imposing evolutionary pressure, we isolated mutations that changed thioredoxin, which is a monomeric disulfide reductase, into a [2Fe-2S] bridged dimer capable of catalyzing O2-dependent sulfhydryl oxidation in vitro. Expression of the mutant protein in Escherichia coli with oxidizing cytoplasm and secretion via the Tat pathway restored disulfide bond formation in strains that lacked the complete periplasmic oxidative machinery (DsbA and Dsb… Show more

“…These enzymatic pathways have been extensively studied in E. coli (reviewed in Ref. 3,58). Functional homologues of Dsb proteins have also been found in Gram-positive bacteria (15)(16)(17)(18)(19)(20)(21)(22); however our knowledge of the processes of oxidative folding in these organisms is scarce.…”

“…Strain GB100 (As MC4100, pcnB1 zad-981::Tn10d (Km r ), dsbA::Cm, ⌬tatABC::aac (3)IV) was constructed from strain MC4100-P (28) as follows. Initially the dsbA::Cm allele from strain LM105 (29) was introduced by P1 transduction. Subsequently, a marked deletion of tatABC was also introduced by P1 transduction.…”

“…These enzymes catalyze thiol-disulfide oxidoreductase reactions that are important in enzyme catalysis and in the maintenance of the correct thiol redox state in proteins. Dsb (disulfide bond) proteins are a specific of TRX-like proteins that are essential for the oxidative folding of secreted proteins in many Gram-negative bacteria (1)(2)(3). In Escherichia coli K-12 five different Dsb proteins catalyze the correct introduction of disulfide bonds in substrate proteins (4).…”
