Introduction: Hepatitis B virus (HBV) constitutes an important risk factor for cirrhosis and hepatocellular carcinoma (HCC). The link between circulating microRNAs and HBV has been previously reported, although not as a marker of liver disease progression in chronic hepatitis B (CHB). The aim of this study was to characterize miRNA expression profiles between CHB with and without cirrhosis or HCC. Methods: A total of 12 subjects were recruited in this study. We employed an Affymetrix Gene Chip miRNA 3.0 Array to provide universal miRNA coverage. We compared microRNA expression profiles between CHB with and without cirrhosis/HCC to discover possible prognostic markers associated with the progression of CHB. Results: Our results indicated 8 differently expressed microRNAs, of which miRNA-935, miRNA-342, miRNA-339, miRNA-4508, miRNA-3615, and miRNA-3200 were up-regulated, whereas miRNA-182 and miRNA-4485 were down-regulated in patients with CHB who progressed to cirrhosis/HCC as compared to those without progression. Conclusions: We demonstrated the differential expression of between patients with HBV without cirrhosis/HCC and those who had progressed to these more severe conditions. These miRNAs may serve as novel and non-invasive prognostic markers for early detection of CHB-infected patients who are at risk of progression to cirrhosis and/or HCC.

Some reports have evaluated the prognostic relevance of microRNAs (miRNAs) in patients with pancreatic cancer (PC). However, most studies focused on limited miRNAs with small number of patients. The aim of the study is to identify a panel of miRNA signature that could predict prognosis in PC with the data from The Cancer Genome Atlas (TCGA). A total of 167 PC patients with the corresponding clinical data were enrolled in our study. The miRNAs significantly associated with overall survival (OS) in PC patients were identified with Cox proportional regression model. A risk score formula was developed to evaluate the prognostic value of the miRNA signature in PC. Thirteen miRNAs were identified to be significantly related with OS in PC patients. Patients with high risk score suffered poor overall survival compared with patients who had low risk score. The multivariate Cox regression analyses showed that the miRNA signature could act as an independent prognostic indicator. In addition, the signature might serve as a predicator for treatment outcome. Our study identified a miRNA signature including 13 miRNAs which could serve as an independent marker in prognosis of PC.

BackgroundSpecific biomarkers for outcome prediction of lung squamous cell carcinoma (LUSC) are still lacking. This study assessed the prognostic value of differentially expressed miRNAs of LUSC patients.ResultsTwelve of the 133 most significantly altered miRNAs were associated with overall survival (OS) across different clinical subclasses of the Cancer Genome Atlas (TCGA) LUSC cohort. A linear prognostic model of seven miRNAs was developed to divide patients into high- and low-risk groups. Patients assigned to the high-risk group exhibited poor OS compared with patients in the low-risk group, which was further validated in the validation cohort and entire LUSC cohort.MethodsMiRNA expression profiles with clinical information of 447 LUSC patients were obtained from TCGA. Most significantly altered miRNAs were identified between tumor and normal samples. Using survival analysis and supervised principal components method, a seven-miRNA signature for prediction of OS of LUSC patients was established. Survival receiver operating characteristic (ROC) analysis was used to assess the performance of survival prediction. The biological relevance of predicted miRNA targets was also analyzed using bioinformatics method.ConclusionsThe current study suggests that seven-miRNA signature may have clinical implications in the outcome prediction of LUSC.
