An evaluation of buffered aspirin and aspirin tablets in postoperative pain after third molar surgery.

Abstract: 1. Single doses (500 and 1000 mg) of both buffered aspirin and aspirin tablets were compared with placebo in a randomised double‐blind trial of parallel design in patients with postoperative pain after third molar surgery. 2. Only buffered aspirin 500 mg provided significant pain relief (P = 0.016) during the 5 h investigation period. 3. A significant correlation (P = 0.004) was observed between overall pain scores after the various aspirin treatments and aspirin esterase activity. 4. Buffered aspirin preparat… Show more

“…There were 17 citations [14–30] with relevant direct comparisons (18 trials) of more than one dose of aspirin. There were four comparisons of 500 and 1000 mg and 10 of 600 or 650 g and 1000 or 1200 mg. Other comparisons included doses of 200, 300, 325, 625, 650, 750 and 900 mg.…”

“…These include formulation, dose and plasma concentration of acetylsalicylate. [2][3][4][5] The latter appears to be determined by plasma aspirin esterase activity. By contrast, there is limited information on the efficacy of paracetamol in postoperative dental pain.…”

“…Intervention In the early postoperative period, patients were medicated with either a single dose of soluble aspirin 900 mg, solid paracetamol 1,000 mg or placebo. Main outcome measures Pain intensity was measured on 100 mm visual analogue scales at 0,5,10, 15, 20, 30, 45, 60, 90, 120 and 240 minutes after dosing. Other efficacy variables evaluated included time to rescue medication and an overall assessment of the study medication efficacy by the patient on completion of the study.…”

“…Especially, for this purpose, older people are commonly medicated with this agent 2. The so-called 'aspirin resistance (or aspirin non-responsiveness) syndrome' is characterized by an incomplete inhibition of platelet function by aspirin 2, 3. It may account for the high variability in effective anti-coagulant doses of aspirin in the population 2, 3.…”

“…The so-called 'aspirin resistance (or aspirin non-responsiveness) syndrome' is characterized by an incomplete inhibition of platelet function by aspirin 2, 3. It may account for the high variability in effective anti-coagulant doses of aspirin in the population 2, 3. Increased hydrolysis of circulating aspirin, pharmacokinetically corresponding to faster elimination of the drug from the circulation by transforming it into salicylate and acetate, is thought to participate as a causal factor in the aspirin resistance 4, 5.…”
