A study was Initiated to determine which preservatives, frequently used in ophthalmic solutlons, bind with SOFLENSQ (polymacon) Contact Lenses. Zone of inhibition assays wwe employed to detect and measure preservatlve binding. Lenses soaked In preservative solutions at concentrations normally used in ophthalmic preparations were subjected to varlous extraction techniques. These lenses were then placed on seeded agar plates of Staphylococcus aureus or Trichophyton mentagrophytes, Incubated and examined for the appearawe of zones of inhibition. The amount of extraction required to prevent zone formation was interpreted as a measure of preservative binding. Results showed that: (1) thlmerosal, thlmerosal with disodlum edetate, and propylparaben bind weakly; (2) phenylmercuric acetate and phenylmercuric nitrate bind moderately; and (3) chlorhexidlne gluconate, benzalkonlum chloride, and an alkyl trlethanol ammonium chloride compound bind strongly to the lens material.

Mo1.-Gew. (Raet) gef. 293,8. Ausbeute Infrarotes Absorptionsspektrum : unter identisohen R.ettktionsbedingungeu wie bei 4 die folgende Substam: 12,M g (W%). V, , 1714 s, 1671 s, 1672 8, 1461 s, 1286 schw, 1246 m, 1191 s (S = 0 Frequenz fur 1 : 3 sulfite), 9& s, 930 schw, 850 (breites Band), 790 m. Zuaamme&Mg Die experimentellen Resultate lassen die folgenden Schlusse zu: Wiihrend der Kondensation von Theophyllin-Kalium mit 3-Chloropropylensulfit findet eine intramolekulare Umlagerung statt, aus welcher die Bildung eines symmetrischen 1 : 3-Sulfitringes resultiert. Dieae Umlagerung hat eine gesunde theoretische Grundlage in den Arbeiten von Winstein und Mitarbeitern, welche von B. R. Alemnderg) vorztiglich zusammengefal3t wurden. Die Bildung des 1:3-Sulfitringes wurde durch unabhiingige Synthese uber 2-Chlorotrimethyleneulfit eindeutig bewiesen. Die Verbindungen, die auf beiden Wegen erhalten wurden, hatten identische Schmelz-und Mischschmelzpunkte sowie ahnliche Absorptionskurven im Infrarot-Spektrum. Absorptionsmaxima, die nicht a d besondere funktionelle GFppen hinweisen, zeigen gute ffbereinatimmung mit Werten, die von anderen Bearbeitern fiir ahnliche 1 : 3-Sulfite gefunden wardens). Diese Substanz (11) wird auf ihre pharmakologischen Eigenschaften gepriift und Endergebnisse werden sodann mitgeteilt werden. Ich bin Prof. H . Heimun, der es mir ermijglichte, in der Abteilung fiir Industrielle Chemie dee Israel Instituts for Technology Haifa diese Arbeit durchzufiihren, zu groBem Dank verpflichtet. AuBerdem danke ich dem Chemistry Department fiir die Infrarot-Spektren und Dr. 8. Fruman von derselben -4bteilung fiir wertvolle Ratschlie hinsichtlich der Bddungsmechanismen.
