An Evolutionarily Conserved SoxB-Hdac2 Crosstalk Regulates Neurogenesis in a Cnidarian

Abstract: SUMMARY SoxB transcription factors and histone deacetylases (HDACs) are each major players in the regulation of neurogenesis, but a functional link between them has not been previously demonstrated. Here, we show that SoxB2 and Hdac2 act together to regulate neurogenesis in the cnidarian Hydractinia echinata during tissue homeostasis and head regeneration. We find that misexpression of SoxB genes modifies the number of neural cells in all life stages and interferes with head regeneration. Hdac2 was co-expresse… Show more

“…Furthermore, neurogenesis occurs in both the ectoderm and endoderm in Nematostella . Hydrozoan i-cells are first found in the endoderm, and embryonic neurogenesis occurs within this tissue (Flici et al, 2017; Martin, 1990), also pointing to a derived mode of neurogenesis in hydrozoans. Co-occurrence of the i-cell lineage, exclusively endodermal embryonic neurogenesis, and the loss of Notch mediated neurogenesis inhibition in hydrozoans are consistent with a link between these phenomena.…”

“…In order to generate the Notch intracellular domain (NICD) overexpression construct, the NICD was amplified from cDNA using primers NICDfwd (5'ATGAAAAGGACCTATGGTGCCC3') and NICDrev (5'GCATTGAAATTATATAGTTCGTGCTACAAC3') and cloned into the β-tubulin ::GFP plasmid using Gibson assembly to generate an NICD-GFP fusion separated by a linker region as used previously (Flici et al, 2017). Injections were carried out as previously described (Millane et al, 2011).…”

“…Given the importance of histone deacetylation in gene regulation and differentiation, it may be a conserved method utilised by many animals to regulate changes to global transcription from the genome during regeneration. Reduced HDAC activity also prevents formation of the blastema in the cnidarian Hydractinia head regeneration by both HDACi and HDAC2 knockdown (Flici and Frank, 2018;Flici et al, 2017). In vertebrates, HDACi prevents hindlimb muscle regeneration in mice (Spallotta et al, 2013), and tail and limb regeneration in Xenopus (Taylor and Beck, 2012;Tseng et al, 2011).…”

“…Most recently, genetic engineering (through both gene knockouts 9 and knockins 10 ) has been achieved in Hydractinia by means of CRISPR/Cas9 embryo microinjection. Prior to the advent of these gene editing techniques, antisense RNA-mediated gene silencing to induce gene-specific knockdowns had been frequently employed in Hydractinia, including morpholino microinjection in embryos 11 and double-stranded RNA (dsRNA) soaking in embryos and polyps 7,[12][13][14][15] . dsRNAs exploit the endogenous RNA interference (RNAi) machinery of the cells and have been used as molecular tools to transiently lower the transcript levels of a particular target gene (i.e., knockdown) for decades 16 .…”
