An Experimental Model for the Study of Lymphedema and its Response to Therapeutic Lymphangiogenesis

Abstract: The post-surgical murine tail model of lymphedema closely simulates attributes of human lymphedema and provides the requisite sensitivity to detect therapeutically induced functional and structural alterations. It can, therefore, be used as an investigative platform to assess mechanisms of disease and its responses to candidate therapies, such as therapeutic lymphangiogenesis.

“…18 In previous studies, administration of VEGF-C to promote therapeutic lymphangiogenesis was shown to reduce edema volume and have beneficial effects on tissue architecture. 19,20 Molecular characterization of the tissues with lymphedema revealed the prominence of inflammatory mediators affecting the inflammatory nature of this acquired microvascular disorder. [21][22][23] Lymph stasis is known to induce the tissue destruction associated with lymphedema.…”

“…Upon examination, treated animals were found to have reversal of the lymphedematous state to the normal state with resolution of edema, hypercellularity, inflammatory changes, and microlymphatic dilation. Both lymphatic vessel number and cross-sectional area were reduced following exogenous administration of the recombinant VEGF-C (Cheung et al, 2006). In another mouse model of chronic obstructive lymphedema, treatment with VEGF-C also improved lymphedema in the studied animals.…”
