An in vivo model for measuring antigen‐induced SRS‐A‐mediated bronchoconstriction and plasma S RS‐ A levels in the guinea‐pig

Abstract: 1 Pharmacological modulation of antigen-induced anaphylaxis in actively sensitized guinea-pigs with intravenously administered indomethacin (10 mg/kg), pyrilamine (2.0 mg/kg) and propranolol (0.1 mg/kg) resulted in a delayed onset, slowly developing bronchoconstriction indicative of a slow-reacting substance of anaphylaxis (SRS-A) response. 2 Measurements of pulmonary mechanics on the drug-pretreated animals challenged with ovalbumin demonstrated a more prominent effect on dynamic compliance than resistance. “…Thus in both cases the predominant mediators responsible for the residual bronchospasm in the presence of anti-histamine and cyclo-oxygenase blockade appear to be leukotrienes, as has been previously found with i.v. challenge (Ritchie et al, 1981;Anderson et al, 1983). It is possible that following a low challenge stimulus (microshock), a redirection of arachidonic acid metabolism (with increased leukotriene production) may occur, whereas following a high challenge stimulus (macroshock) and lipoxygenase pathway is already maximally stimulated.…”

“…challenge in the alation of presence of a histamine H,-receptor antagonist (Rit-1987). This chie et al, 1981;Anderson et al, 1983). In the latter uantitative situation, indomethacin had been used to enhance in airway histamine-independent resistant bronchospasm.…”

“…Th plasma supernatant was then decanted into a test tube, extracted with 4 volumes of absolute ethanol, and allowed to stand on ice for 30 min. The precipitated protein was removed by centrifugation at 25,000 x g for 30 min at 4°C (Anderson et al 1983). The ethanol supernatant was evaporated under vacuum at 35°C.…”
