“…After the run, the gel was removed and washed with 2.5% (v/v) Triton X-100 in 100 mM Tris-HCl buffer pH 8.5. After this step, the gel slab was layered onto a 4.5% polyacrylamide gel slab containing 1% gelatin and left for 2 h at 30 1C, essentially as described by Jorda˜o et al (1996). Finally, the original gel slab was removed and the gelatincontaining gel slab was stained with 0.1% (w/v) Coomassie Blue R-250 in water-ethanol-acetic acid (5:5:2 by volume) for at least 6 h. “…In Stomoxys , the regulation of digestive enzymes have been shown to be at the posttranscriptional level with protein levels reaching a maximum shortly after obtaining the blood meal (8 -10 h) (Mofatt et al , 1995). The opaque zone cells are found to contain many vesicles where the digestive enzymes are stored as zymogens and are released quickly to the gut lumen in response to the blood meal (Jordao et al , 1996). Two gut-specific cDNA products have been molecularly characterized from Stomoxys ( Ssp1 and Ssp2 ) and both are found to contain serine protease motifs with chymotrypsin and trypsin specificities, respectively (Lehane et al , 1998).…”
