An improved Huffman coding method for archiving text, images, and music characters in DNA

Abstract: An improved Huffman coding method for information storage in DNA is described. The method entails the utilization of modified unambiguous base assignment that enables efficient coding of characters. A plasmid-based library with efficient and reliable information retrieval and assembly with uniquely designed primers is described. We illustrate our approach by synthesis of DNA that encodes text, images, and music, which could easily be retrieved by DNA sequencing using the specific primers. The method is simple … Show more

“…The potential of DNA has been realized for various information-theoretic protocols, including DNA steganography for the identification of genetically modified organisms [12,21,23] hiding of messages in DNA [9,11,16,20] and long-term data storage in DNA [3,7,8,10,14,21]. These applications require mechanisms to validate, ensure, and possibly verify message accuracy.…”

“…Advancement in using DNA as an information storage medium continues, both experimentally (Pak Chung Wong, 2003; Kashiwamura et al, 2005; Yachie et al, 2007) and algorithmically (Ailenberg and Rotstein, 2009). Yet, practical and large-scale implementation remains unfavorable due to technological limitations on robust DNA synthesis and sequencing technology.…”
