An inhalation anaesthesia approach for neonatal mice allowing streamlined stereotactic injection in the brain

Abstract: Highlights Development of inhalation-based anaesthesia for early postnatal (P0−2) mice. 3D-printed mould allows anaesthetic maintenance for neonatal surgery. Improved mouse welfare through reliable neonatal inhalation anaesthesia. Rapid procedure for brain transduction of mouse litter in under 2 h.

“…In vivo, the viral injection was performed according to 76 . P1 neonatal mice were anaesthetised using isoflurane, bilaterally injected using a pulled glass needle in the hippocampal area with 0.5 μL AAV-hSyn-Cre-EGFP at 3 × 10 12 GC ml −1 (Addgene #105540, virus produced by U -Penn Vector Core), and returned to the parent cage until P7, when tissue was harvested for organotypic culture.…”

“…Adenovirus 9 (AAV9)‐based overexpression vector of TGIF2 (AAV9‐TGIF2) and the empty vector (AAV9‐NC) were procured from Genechem Co., Ltd (Shanghai, China). The AAV9‐based vectors were injected into the newborn mice (P1) in the VPA group intracerebroventricularly according to a previous report (Ho et al., 2020 ). The injection was performed using a small animal stereotactic frame, and the virus was delivered using Hamilton syringes and microinjection adaptors.…”

“…Wait for loss of motor response. Once anesthetized, transfer the pup to the heating pad and place the snout into a 3D-printed whole-body mouse mold (we use a mold from a previously published protocol 7 ). Keep the mouse under anesthesia (oxygen 1 L/min, isoflurane 4%).…”
