An Integral Role for Heme Oxygenase-1 and Carbon Monoxide in Maintaining Peripheral Tolerance by CD4+CD25+ Regulatory T Cells

Abstract: Over the past decade, a great deal of interest and attention has been directed toward a population of regulatory T cells (Treg) coexpressing the markers CD4 and CD25. The hallmark phenotype of this cell population resides in its ability to dominantly maintain peripheral tolerance and avert autoimmunity. Despite robust research interest in Treg, their mechanism of action and interaction with other cell populations providing immune regulation remains unclear. In this study, we present a model for Treg activity t… Show more

“…IDO catalyzes the rate-limiting step of tryptophane degradation. Kynurenine, one of the products of this reaction, affects proliferation as well as differentiation of helper T cells (Brusko et al, 2005;Munn and Mellor, 2007). Likely, in an inflammatory environment the presence of AC might help to promote healing, once an inflammatory stimulus is eliminated, by modulating adaptive immune responses.…”

“…21 This has led to the proposal that HO-1 and CO may mediate immune tolerance by favoring the induction of master immunosuppressor cells, the so-called regulatory T cells (Tregs). 22,23 Therefore, the activation of HO-1 expression can be thought of as a critical parameter to switch the classical proinflammatory activity of macrophages into an immunoregulatory one. It remains to be determined whether, at high concentrations of intracellular heme, as encountered after transfusion of stored blood, the HO system becomes overwhelmed and therefore is unable to neutralize the oxidative and inflammatory effects.…”

“…64 Inhibition of neutrophil aggregation also may underlie the anti-inflammatory effects of CO. 65 CO also increases regulatory T cells, which inhibit the Thelper 1 (Th1) cytokine response. 66 Due to its potential toxicity, CO has not been trialled in human subjects.…”
