An ischemia-homing bioengineered nano-scavenger for specifically alleviating multiple pathogeneses in ischemic stroke

Abstract: Background Ischemic stroke is one of the most serious global public health problems. However, the performance of current therapeutic regimens is limited due to their poor target specificity, narrow therapeutic time window, and compromised therapeutic effect. To overcome these barriers, we designed an ischemia-homing bioengineered nano-scavenger by camouflaging a catalase (CAT)-loaded self-assembled tannic acid (TA) nanoparticle with a M2-type microglia membrane (TPC@M2 NPs) for ischemic stroke … Show more

“…Cerebrovascular diseases are common in neurology and are global public health issues. 16 According to the latest statistics, cerebrovascular diseases have ranked first in the death cause spectrum of urban and rural residents in China 17 and second around the world. 18 In recent years, the widely used neuroelectrophysiological techniques, such as SSR, Somatosensory evoked potential (SEP), and Motor evoked potential (MEP), can sensitively display brain dysfunctions as well as small or subclinical lesions that cannot be found by imaging examinations.…”

“…A new trend for microglia immunomodulation involves the functionalization of NPs through microglia components, as reported by Duan et al. [ 270 ] in the context of ischemic stroke treatment ( Figure ). They designed a nano‐scavenger composed of a self‐assembled core containing tannic acid (TA) and the enzyme CAT with a cell membrane coating derived from M2‐polarized microglia.…”

“…Reproduced with permission. [270] M2 phenotype at both mRNA and protein levels, suppressing pro-inflammatory responses and exhibiting a neuroprotective effect. [274] Promising results about the use of AuNCs have also been observed in treating SCI in combination with berberine, a natural anti-inflammatory quaternary ammonium alkaloid isolated Coptidis rhizoma, demonstrating neuronal apoptosis alleviation by promoting M2 polarization.…”

“…[94] In recent years, nanotherapeutics, utilizing nanotechnology to alter the pharmacology of drugs and therapies, show great potential in revolutionizing the treatment of many diseases including IBD and other inflammatory diseases owing to their advantages such as targeted drug delivery, improved drug solubility, extended drug half-life, and reduced drug immunogenicity. [95][96][97][98][99][100] In particular, small-molecule drugs with intrinsic antioxidant and anti-inflammatory effects are the first choice to alleviate oxidative stress in IBD. However, the poor water solubility greatly limits their clinical applications.…”
