Background: The organophosphorus (OP) pesticide malathion is a highly neurotoxic compound, leading to cholinergic syndrome. Malathion produces reproductive toxicity. Objectives: To investigate the protective effects of curcumin on the testicular toxicity induced in male mice orally administrated with malathion.Materials and methods: Forty-eight mature male mice were used. The mice were divided into four groups, 12 mice each, and treated for 4 weeks as follows: the control (G1), curcumin-treated group (G2), malathiontreated group (G3), and the curcumin plus malathion-treated group (G4). At the end of the experiment, the mice were sacrificed and dissected, and tissue samples were obtained and antioxidant parameters were examined. Tissue samples were also histopathologicaly and immunohistochemically assessed. Samples of blood were collected for assessing the levels of serum hormones, namely follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone, and acetylcholinesterase activity (AChE). Results: Malathion exposure caused a decrease in serum testosterone and LH levels and antioxidant enzymes (SOD and CAT) and elevation of oxidative stress marker (MDA). Histological changes were as well observed in the form of maturation abnormalities, intratubular necrosis, and inflammatory infiltrate in malathion-treated mice. Co-administration of curcumin with malathion reduced lipid peroxidation (LPO) activity and improved spermatogenesis and increased catalase (CAT), superoxide dismutase (SOD), and testosterone levels. Conclusion:The current study showed that curcumin could ameliorate the elevated concentration of lipid peroxidation products, increase the levels of antioxidative markers, and improve the pathological changes in the testis induced by malathion administration. The capacity of curcumin to work as antioxidant supposedly relates to its structure.

Glioblastoma multiforme (GBM), a grade IV astrocytoma, is the most common and deadly type of primary malignant brain tumor, with a patien’s median survival rate ranging from 12 to 15 months. Over the last fifteen years, the treatment for GBM has included maximal safe surgical resection with combination radiotherapy and adjuvant temozolomide chemotherapy. The low efficacy of mentioned therapies has forced researchers to explore an appropriate alternative or complementary treatment for GBM. It has been shown that curcumin has therapeutic potentials to fight against GBM via affecting on cell proliferation, apoptosis, cell cycle, invasion and angiogenesis pathways. In addition, curcumin possess a synergistic impact with chemotherapeutic agents. Herein, we summarized the current findings on curcumin as potential therapeutic agent in the treatment of GBM.

Modulatory effects of curcumin on the atherogenic activities of inflammatory monocytes: Evidence from in vitro and animal models of human atherosclerosis

Atherosclerosis is a complex and long‐lasting disorder characterized by chronic inflammation of arteries that leads to the initiation and progression of lipid‐rich plaques, in which monocytes/macrophages play the central role in endothelial inflammation and taking up these lipids. Circulating monocytes can adopt a long‐term proinflammatory phenotype leading to their atherogenic activities. During atherogenic condition, inflammatory monocytes adhere to the surface of the activated endothelial cells and then transmigrate across the endothelial monolayer into the intima, where they proliferate and differentiate into macrophages and take up the lipoproteins, forming foam cells that derive atherosclerosis progression. Therefore, modulating the atherogenic activities of inflammatory monocytes can provide a valuable therapeutic approach for atherosclerosis prevention and treatment. Curcumin is a naturally occurring polyphenolic compound with numerous pharmacological activities and shows protective effects against atherosclerosis; however, underlying mechanisms are not clearly known yet. In the present review, on the basis of a growing body of evidence, we show that curcumin can exert antiatherosclerotic effect through inhibiting the atherogenic properties of monocytes, including inflammatory cytokine production, adhesion, and transendothelial migration, as well as intracellular cholesterol accumulation.
