An Lnc RNA (GAS5)/SnoRNA-derived piRNA induces activation of TRAIL gene by site-specifically recruiting MLL/COMPASS-like complexes

Abstract: PIWI-interacting RNA (piRNA) silences the transposons in germlines or induces epigenetic modifications in the invertebrates. However, its function in the mammalian somatic cells remains unknown. Here we demonstrate that a piRNA derived from Growth Arrest Specific 5, a tumor-suppressive long non-coding RNA, potently upregulates the transcription of tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL), a proapoptotic protein, by inducing H3K4 methylation/H3K27 demethylation. Interestingly, the P… Show more

“…[710] Experimental manipulation of PIWI/piRNA expression both in vitro and in vivo influences growth, invasion and migration of the cancer cells. [7, 1113] In addition, recent studies have shown a possible role of piRNAs in somatic cells, including the regulation of protein coding genes. [1416] piRNAs can act in this context similarly to microRNAs by binding to the 3'UTR of messenger RNA and inducing its degradation.…”
